妊娠期糖尿病孕妇胎盘组织中脂联素的表达及滋养细胞增殖的变化 被引量:4

摘要 目的探讨妊娠期糖尿病孕妇胎盘组织中脂联素的表达及滋养细胞增殖的变化情况。方法选择2011年11月至2012年1月在中山大学附属第一医院进行产前检查并行择期剖宫产的30例妊娠期糖尿病孕妇为研究组,30例正常糖耐量孕妇为对照组。分别采用实时荧光定量聚合酶链反应、Western印迹和免疫组织化学3种方法检测研究组及对照组胎盘组织中脂联素的表达水平,采用免疫组织化学方法检测增殖细胞核抗原阳性细胞数。比较2组胎盘滋养细胞的增殖情况;比较2组年龄、孕周等临床资料和新生儿出生体重等指标,并追踪其子代的生长发育情况。组间比较采用两独立样本t或x^2检验。结果研究组平均年龄为(32.3±4.4)岁,高于对照组的(29.2±4.1)岁(t=2.78,P〈0.01);胎盘重量为(0,7±0.1)b,高于对照组的(0.6±0.1)kg(t=5.50,P〈0.01);新生儿平均出生体重为(3.4±0.4)kg,高于对照组的(3.2±0.3)kg(t=2.28,P〈0.05);但2组新生儿头围、身长等指标差异无统计学意义,出生后1和3个月的体重、身长差异亦无统计学意义(P值均〉0.05)。研究组胎盘组织中脂联素mRNA和蛋白表达水平分别为1.18±0.12和0.93±0.19,低于对照组(2.40±0.23和1.44±0.29)(t值分别为25.61和8.06,P值均〈0.01),增殖细胞核抗原阳性细胞数为(1056.7±127.8)个,高于对照组的(387.2±87.1)个(t=-14.48,P〈0.01)。结论妊娠期糖尿病孕妇胎盘组织中脂联素表达水平下降,滋养细胞增殖增加,但对子代生长发育的影响尚不明确。
