摘要
本试验利用体外培养的未成熟仔猪睾丸支持细胞,应用实时荧光定量PCR研究了FSH对CyclinA2 mRNA表达的影响。结果发现促卵泡素(FSH)以时间和浓度依赖的方式促进了CyclinA2 mRNA的表达(P<0.05);Forskolin促进了支持细胞CyclinA2 mRNA的表达,而Rp-cAMP、Verapamil和U0126都抑制了其表达(P<0.05),但这3种抑制剂单独作用时对CyclinA2 mRNA的表达与对照相比无显著差异(P>0.05)。这表明FSH以浓度和时间依赖的方式诱导了CyclinA2 mRNA的表达,并通过cAMP、Ca2+内流和细胞外信号调节的蛋白激酶1/2(ERK1/2)通路调节仔猪睾丸支持细胞CyclinA2 mRNA的表达。
In the present study,cultured immature boar Sertoli cells were used to study the effect of cAMP production,Ca2+in-flux and ERK1/2 pathway on the relative expression of CyclinA2 mRNA using Real-time RT-PCR. Our result showed that FSH af-fected the expression of CyclinA2 mRNA time and dose dependently(P0.05);Forskolin facilitated the expression of CyclinA2 mRNA in the FSH-induced Sertoli cells,while Rp-cAMP,Verapamil,and U0126 suppressd the expression of CyclinA2 mRNA(P0.05).However,these inhibitors had no significant effect on the expression of CyclinA2 mRNA, compared to the control when theywere used alone(P0.05). These results inferred that FSH might regulate the expression of CyclinA2 mRNA in cultured immatureboar Sertoli cells mainly through affecting the cAMP production, Ca2+influx and activation of ERK1/2 pathway.
