慢性牙周炎患者血清miRNA-146a的表达及其靶基因预测分析 被引量:2

摘要 目的:探讨慢性牙周炎患者血清miRNA-146a的表达情况及其与慢性牙周炎发生发展的可能关系。方法:运用QRT-PCR方法检测miRNA-146a在慢性牙周炎患者血清和健康对照组血清的表达情况,分析其与临床病理参数的关系;运用TargetScan靶基因预测分析软件等生物信息学方法,预测miRNA-146a靶基因,筛选与慢性牙周炎密切相关基因。结果:miRNA-146a在慢性牙周炎患者血清和健康对照组血清的2-△△CT值分别为4.6567±4.3296、0.7748±0.7840,与健康对照组相比,miRNA-146a在慢性牙周炎患者血清高表达(P<0.05);miRNA-146a的表达水平与性别、年龄没有明显关系(P>0.05),与临床分期有关,miRNA-146a随着疾病的加重而增高(P<0.05)。生物信息学分析显示TLR4、CTSC、MPO、COL1A1、HSPG2、HLA-C为慢性牙周炎中miRNA-146a的靶基因。结论:miRNA-146a在慢性牙周炎患者血清中高表达,可能通过调控其下游靶基因,在慢性牙周炎发生过程中起着重要作用。 Objective: To investigate the expression and clinical significance of miRNA-146a in serum of chronic periodontitis patients. Methods: The miRNA-146a in serum of patients with chronic periodontitis and healthy controls were analyzed by quantitative real-time PCR.The relationship between the expression of miRNA-146a and clinical parameters was analyzed. Target genes of miRNA-146a were predicted with bioinformatics methods such as TargetScan and closely related genes with Periodontal disease were screened. Results: Compared with healthy controls, miRNA-146a was significantly increased(P〈0.05); Bioinformatics analysis revealed that TLR4、 CTSC、 MPO、 COL1A1、 HSPG2、 HLA-C might be the target genes of miRNA-146a in chronic periodontitis. Conclusion: Our results demonstrated that altered miRNA-146a expression may be involved in the pathogenesis of chronic periodontitis through regulation of its target genes.
