摘要
目的观察回药复方软化动脉胶囊对糖尿病(Diabets Mellitus,DM)并发动脉粥样硬化(atherosclerosis,AS)大鼠主动脉形态学与MMP-9基因表达的影响。方法采用高脂饲料喂养和STZ腹腔注射(45mg/kg)的方法建立糖尿病并发动脉粥样硬化大鼠模型。给药12周后,各组大鼠主动脉病理切片观察组织形态学变化,采用实时荧光定量PCR技术检测主动脉MMP-9m RNA的水平。结果回药复方软化动脉胶囊可以改善糖尿病并发动脉粥样硬化大鼠的主动脉的病理改变,抑制主动脉MMP-9m RNA的表达(P<0.05)。结论回药复方软化动脉胶囊可以改善糖尿病并发动脉粥样硬化大鼠主动脉的病理改变,延缓糖尿病并发动脉粥样硬化病变的发展,其机制可能与抑制主动脉MMP-9 m RNA的表达有关。
Objective to investigate Huiyao Compound Ruanhua Dongmai Capsule's effect on artery morphology and aorta matrix metallo proteinase9(MMP9) m RNA expression in diabetes and atherosclerosis model rats. Methods Rats of diabetes with atherosclerosis were modeled by STZ( 45mg/kg) and high fat diet. After twelve weeks of treatment, artery morphology was observed and real-time quantitative PCR technology was employed to measure MMP9 m RNA in aorta. Results Huiyao Compound Ruanhua Dongmai Capsule could improve pathological changes and inhibite the in aorta MMP9 m RNA 's expression in diabetes and atherosclerosis nodel rats( P〈0.05). Conclusion Huiyao Compound Ruanhua Dongmai Capsule has a good effect on delaying the development of diabetes and atherosclerosis. The mechanism is probably related to its effect of inhibiting the expression of MMP9 in aorta.
