摘要
该实验研究了蜂毒肽在不同溶剂(水、脱氧水、生理盐水、磷酸缓冲盐溶液、50%乙醇、乙醇、丙三醇)中的稳定性,发现蜂毒肽在50%乙醇中较在水、脱氧水、生理盐水、磷酸缓冲盐溶液中稳定,与乙醇、丙三醇相比,可完全溶解蜂毒中的蜂毒肽,故选择50%乙醇作为蜂毒肽短期储备的储存溶剂;考察了蜂毒肽受光照、温度、p H的影响,以及在不同浓度磷酸缓冲盐溶液、不同p H磷酸缓冲盐溶液和大鼠皮肤匀浆液中的稳定性,结果表明蜂毒肽不受光照、温度、p H的影响,在低浓度、低离子强度溶液中降解加快,在磷酸缓冲盐溶液中随着p H升高降解加快,在大鼠皮肤匀浆液中加速了降解,为选择影响蜂毒肽稳定性小的实验条件提供了实验依据。
The stability of melittin in different solvents( water,deoxygenated water,physiological saline,PBS,50% ethanol,ethanol,glycerol) was studied and the results showed that the stability of melittin is not influenced by light,temperature and p H in50% ethanol,which melittin can be completed dissolved when compared with ethanol and glycerol,in such,50% ethanol was chosen as solvent storage when measured content of melittin. Then the effect of different concentrations of PBS,the p H of PBS and rat skin homogenates were tested,and the results showed that melittin was degraded rapidly at low concentration solution and low ionic strength.Increasing p H of PBS and rat skin homogenate can accelerate the degradation of melittin. These researches provide an experimental basis for further study of melittin.
