摘要
目的:建立RP-HPLC-PDA测定石见穿不同部位中5个三萜酸(坡模酸、山楂酸、科罗索酸、齐墩果酸和熊果酸)的定量方法。方法:采用Kromasil C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-水(90∶10,体积比),等度洗脱,检测波长205 nm,流速0.8 m L·min^-1,柱温28℃。结果:坡模酸、山楂酸、科罗索酸、齐墩果酸和熊果酸线性范围分别为0.096 00-2.400,0.123 0-3.075,0.242 0-6.050,0.283 0-7.075和0.273 0-6.825μg(r=0.999 8,0.999 7,0.999 9,0.999 5,0.999 9),平均加样回收率(n=6)分别为98.43%,98.13%,100.6%,98.19%,99.15%(RSD 1.3%,0.67%,1.2%,0.87%,0.43%)。结论:该方法操作简便,重复性好,可用于石见穿中主要三萜酸的定量分析。
Objective: To establish a RP-HPLC-PDA method for determination of five triterpenic acids( pomolic acid,hawthorn acid,corosolic acid,oleanolic acid and ursolic acid) in different parts of Salvia chinensis. Method: The isocratic elution and separation was achieved on a Kromasil C18column( 4. 6 mm × 250 mm,5 μm),using acetonitrile-water( 90∶ 10) as the mobile phase at a flow rate of 0. 8 m L·min^-1. The detection wavelength and column temperature were set at 205 nm and 28 ℃,respectively. Result: The calibration curves of pomolic acid,hawthorn acid,corosolic acid,oleanolic acid and ursolic acid were linear over the ranges of 0. 096 0-2. 400,0. 123 0-3. 075,0. 242 0-6. 050,0. 283 0-7. 075 and 0. 273 0-6. 825 μg( r = 0. 999 8,0. 999 7,0. 999 9,0. 999 5,0. 999 9),respectively. The average recoveries were 98. 43%,98. 13%,100. 6%,98. 19% and 99. 15% %,respectively,with RSD( n = 6) being 1. 3%,0. 67%,1. 2%,0. 87% and 0. 43%. Conclusion: The proposed method is so simple and highly reproducible that it promises to be applicable for determination of major triterpenic acids in S. chinensis.
