三首活血化瘀方对气虚血瘀模型大鼠红细胞膜蛋白组成影响的研究 被引量:3

摘要 目的研究补阳还五汤、少腹逐瘀汤和丹参饮对气虚血瘀型大鼠红细胞膜蛋白组成的影响。方法 80只Wistar大鼠,随机分为8组,分别为空白对照组、气虚血瘀模型组、补阳还五汤高低剂量组、少腹逐瘀汤高低剂量组、丹参饮高低剂量组,每组10只。大鼠采取饥饿加力竭游泳法复制气虚血瘀模型,每日1次,连续14天。末次造模后禁食不禁水24 h。每日1次灌胃给药,连续给药15天。取血制备红细胞膜,采用SDS-PAGE法分析红细胞膜蛋白组成变化。结果与空白组比较,模型组带5及带6蛋白含量显著降低(P<0.05)。与模型组比较,补阳还五汤可显著升高带5蛋白(P<0.05)、带6蛋白(P<0.05)和带7蛋白含量(P<0.05);少腹逐瘀汤可显著升高带2蛋白(P<0.05)、带3蛋白(P<0.05)、带5蛋白(P<0.01)和带6蛋白含量(P<0.05);丹参饮可显著升高带4.1蛋白(P<0.05)、带5蛋白(P<0.05)、带6蛋白(P<0.05)和带7蛋白含量(P<0.05)。结论补阳还五汤、少腹逐瘀汤和丹参饮可以不同程度地纠正气虚血瘀模型大鼠红细胞膜的膜蛋白组成。 Objective To investigate the effects of "Buyang Huaiwu Decoction", "Shaofu Zhuyu Decoction" and "Danshen Drink" on changes in the components of erythrocyte membrane proteins of the cold stagnation and blood stasis rats. Methods Eighty Wistar rats were randomly assigned to blank control group, model group and BYHWI) high-dose group and low-dose group, SFZYD high-dose group and low-dose group, DSY high-dose group and low-dose group, with 10 rats in each group. The models were duplicated by starvation and exhausting swimming for 14 days, once a day. After modeling, rats were fed with water and fasted for 24 hours. Rats were treated by decoctions by intragastric administration once a day for 15 days continuously. The composition of erythrocyte membrane proteins was analyzed by SDS-PAGE. Results Compared with the blank group, the percentage composition of band 5 protein and band 6 protein were decreased significantly (P 〈0.05). Compared with the model group, BYHWD could increase the percentage of band 5 protein (P 〈 0.05), band 6 protein ( P 〈 0.05 ) and band 7 protein ( P 〈 0.05), SFZYD could increase the percentage of band 2 protein (P 〈 0.05 ), band 3 protein (P 〈 0.05 ), band 5 protein ( P 〈 0.01 ) and band 6 protein ( P 〈 0.05 ). DSY could increase the percentage of band 4.1 protein (P 〈0.05), band5 protein (P 〈 0.05 ), band 6 protein (P 〈 0.05 ) and band 7 protein (P 〈 0. 05 ). Conclusion BYHWD, SFZYD and DSY could change the composition of membrane protein with different degrees.
