畲药地稔不同时间、部位的没食子酸与槲皮素含量测定 被引量:12

Determination of Gallic Acid and Quercetin in Different Parts and Period of Traditional She Medicine-Melastoma Dodecandrum

摘要 目的通过研究畲药地稔不同部位没食子酸、槲皮素含量的变化规律,确定地稔最佳采收期,研究其全草的药用价值。方法应用高效液相色谱法测定,色谱柱为Agilent-SB C18(4.6 mm×250 mm,5μm);流动相为甲醇-0.4%磷酸溶液,梯度洗脱;流速为1.0 m L·min?1,柱温为30℃;检测波长为254 nm。结果畲药地稔各部位中均含有没食子酸、槲皮素,但是不同月份不同部位两活性成分含量存在差异。结论畲药地稔叶中没食子酸、槲皮素含量均高于茎、根中两成分含量,茎、根中两成分含量无明显区别,地稔全株均有一定的药用价值,以没食子酸、槲皮素含量为指标,从保护中药资源的角度考虑,建议以地上部位入药。最佳采收期在5月初、花未开时。 OBJECTIVE To discover the medicinal value and optimal harvest time of traditional She Medicine-Melastoma dodecandrum by studying the dynamic change of the content of gallic acid and quercetin in different parts of the herb. METHODS HPLC was applied with the chromatographic condition as follows: The chromatographic column was Agilent-SB C18(4.6 mm×250 mm, 5 μm) at 30 ℃; the mobile phrase was methanol-0.4% phosphoric acid, gradient elution; flow rate was 1.0 m L·min^-1; the detection wavelength was 254 nm. RESULTS Gallic acid and quercetin existed in all parts of Melastoma dodecandrum. However, the content of active component contents in different parts varied obviously in different months. CONCLUSION The content of gallic acid and quercetin in leafs is more than stems and roots, but no content difference shows in stems and roots. Taking both gallic acid and quercetin as indices, using the parts on the ground is better for protecting traditional Chinese medicine resources. Moreover, the optimal harvest time should be in early May before flowering.
