双水相萃取长鱼蛋白酶的工艺研究

摘要 采用聚乙二醇(PEG)/盐双水相体系以分离纯化长鱼蛋白酶。通过研究不同成相盐及其浓度、不同PEG分子量及其浓度和不同p H对双水相萃取长鱼蛋白酶的影响。结果表明,最优化的双水相体系为25%PEG1000、20%(NH4)2SO4和p H7.0,在此条件下长鱼蛋白酶上相酶活回收率和纯化倍数分别为90.5%和4.40。 Protease was extracted and purified by the polyethylene glycol(PEG) -salt aqueous two- phase systems from Monopterus albus.Effects of different kinds and concentrations of phasing salts, different molecular weight and concentrations of phasing PEG, pH on extraction of the protease were studied.The results showed that the optimum conditions for extraction and purification of the protease were 25% PEG1000,20% ( NH4)2SO4and pH7.0. Under the optimum extraction conditions, recovery and purification fold of the protease in top phase were 90.5% and 4.40, respectively.
