The Role of Annexin A2 in Carcinogenesis and Progression of Cancer

摘要 膜联蛋白A2是一种钙依赖性蛋白,它在各种类型肿瘤中起正向调节作用,在调节细胞功能上亦扮演多个角色,包括血管生成、增殖、凋亡、细胞迁移、入侵和粘附。膜联蛋白A2与血纤溶酶原以及细胞表面的组织纤溶酶原激活物结合,导致纤溶酶原转化成血纤维蛋白溶酶。血纤维蛋白溶酶是一种丝氨酸蛋白酶,可以激活基质金属蛋白酶和降解细胞外基质,对癌症转移发挥着关键作用。研究发现膜联蛋白A2在卵巢癌细胞中表达增多,能促进癌细胞转移。越来越多的证据表明,膜联蛋白A2及其结合蛋白在恶性肿瘤微环境中发挥重要作用,共同促进癌症的转移。本文综述了膜联蛋白A2及其结合蛋白在恶性肿瘤包括卵巢癌中的生物学作用。 Annexin A2 is a calcium-dependent protein. It is up-regulated in various tumor types and plays multiple roles in regulating cellular functions, including angiogenesis, proliferation, apoptosis, cell migration,invasion and adhesion. Annexin A2 binds with plasminogen and tissue plasminogen activator on the cell surface,which leads to the conversion of plasminogen to plasmin. Plasmin is a serine pro- tease which plays a key role in the activation of metalloproteinases and degradation of extracellular matrix components essential for metastatic progression.The study found that expression of Annexin A2 increased in ovarian cancer cells, and could promote cancer metastasis.Accumulating evidence suggests that interactions between Annexin A2 and its binding proteins play an important role in the tumor mi- croenvironment and act together to enhance cancer metastasis.
