葡萄糖、维生素C浸泡对普安银鲫胚胎发育中乙酰辅酶A羧化酶、脂肪酸合成酶及肉毒碱棕榈酰转移酶Ⅰ活性的影响 被引量:6

Effects of Glucose and Vitamin C Inhabitation on Activities of Acetyl-Co A Carboxylase,Fatty Acid Synthase and Carnitine Palmitoyltransferases Ⅰ during Embryo Development of Carassius auratus gibelio

摘要 为了探究普安银鲫(Carassius auratus gibelio)胚胎发育中乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FAS)和肉毒碱棕榈酰转移酶Ⅰ(CPTⅠ)活性变化及葡萄糖、维生素C浸泡对它们的影响,设置不同浓度的葡萄糖溶液和维生素C溶液用于普安银鲫的孵化,葡萄糖浓度分别为0、5、10、15和20 g/L,维生素C浓度分别为0、20、25、30和35 mg/L。记录出膜时间及孵化率,筛选适宜葡萄糖和维生素C浓度。采用无添加(对照组)、最适葡萄糖浓度(葡萄糖组)和最适维生素C浓度(维生素C组)的溶液用于孵化,测定胚胎发育中ACC、FAS和CPTⅠ活性变化特点。结果表明:1)葡萄糖浓度为15 g/L,维生素C浓度为30 mg/L时能获得最短出膜时间和最高的孵化率。2)胚胎发育过程中,FAS、ACC和CPTⅠ比活力和全活力均呈上升趋势。3)葡萄糖组在原肠中期、晶体出现期和出膜前期ACC和FAS比活力和全活力均显著高于对照组(P<0.05),CPTⅠ比活力和全活力在晶体出现期和出膜前期显著高于对照组(P<0.05)。4)维生素C组ACC、FAS全活力在出膜前期均显著高于对照组(P<0.05)。结果提示,15 g/L葡萄糖和30 mg/L维生素C溶液浸泡能促进普安银鲫胚胎发育过程中ACC、FAS和CPTⅠ的合成与分泌而形成新的代谢水平,以维持胚胎中脂质代谢的动态平衡。 In order to study the changes of activities of acetyl-Co A carboxylase( ACC),fatty acid synthase( FAS),and carnitine palmitoyltransferase Ⅰ( CPTⅠ),and the effects of glucose and vitamin C inhabitation on them during embryo development of Carassius auratus gibelio( C. auratus),glucose solution and vitamin C solutions with different concentrations were used for hatching. The concentrations of glucose were 0,5,10,15 and 20 g / L,respectively,and the concentrations of vitamin C were 0,20,25,30 and 35 mg / L,respectively. Membrane break time and hatching rate were recorded to decide the optimal concentrations of glucose and vitamin C. Solutions without addition( control group) and with optimal concentrations of glucose( glucose group) or vitamin C( vitamin C group) were used for hatching,and the characteristics of changes of ACC,FAS and CPT Ⅰ activities were analyzed during embryo development. The results showed as follows: 1) membrane break time was the shortest,and hatching rate was the highest when the concentrations of glucose and vitamin C were 15 g / d and 30 mg / L,respectively. 2) The specific activity and total activity of ACC,FAS and CPT Ⅰ showed increasing tends during embryo development of C. auratus. 3) The specific activities and total activities of ACC and FAS in glucose group were significantly higher than those in control group at mid-gastrul,crystal appear and prehatching stages( P〈 0.05),and the specific activity an total activity of CPT Ⅰ was significantly higher than that in control group at crystal appear and prehatching stages( P〈 0.05). 4) The total activities of ASS and FAS in vitamin C group were significantly higher than those in control group( P〈 0.05).In conclusion,the inhabitation in solutions with appropriate concentrations of glucose( 15 g / L) and vitamin C( 30 mg /L) can promote synthesis and secretion of ACC,FAS and CPTⅠ during embryo development of C.auratus,and form newmetabolic levels to maintain the dynamic balance of lipid metabolism in embryo.
