摘要
目的:研究植入用顺铂兔局部给药后生物样品中药物浓度的测定方法,并分别测定游离铂及总铂的浓度。方法:改进石墨炉原子吸收法的升温程序和上样方法;增加生物样品的超滤前处理方法,以去除顺铂的蛋白结合物便于测定游离铂含量;并按《中华人民共和国药典》的规定对测定方法进行验证和评估。结果:本测定方法的回收率为80%-110%,日内精密度(RSD)在5%以下,均符合《中华人民共和国药典》2010年版规定。组织中游离铂浓度约为总铂浓度的10%-30%,静脉血中游离铂浓度是总铂浓度的5%以下。结论:当前建立的石墨炉原子吸收方法能较好地测定出血液和组织样本中的游离铂和总铂浓度,且该方法无爆沸现象、重现性好、灵敏度高、数据准确。
Objective: To determine the free and total platinum levels in biological samples from rabbits treated by cisplatin implant administration. Methods: In this experiment, an improved methods for temperature- programming and loading were introduced, and a uhrafihration technology for removing cisplatin-protein conjugates from biological samples was studied. Then, the measurement method for free and total platinum levels was analyzed and evaluated according to the stands of Chinese Pharmacopoeia. Results: After using this method, the recovery was 80% - ll0% and the intraday precision (RSD) was below 5% , which were entirely consistent with the regu- lations of the Chinese Pharmacopoeia 2010 edition. In tissues, the free platinum level accounted for approximately 10% -30% of the total platinum level, and in the venous blood, the free platinum level was even below 5%. Conclusion : The method for the determination of free and total platinum levels in biological samples by atomic fluo- rescence spectrometry has been developed. The method has good reproducibility, high sensitivity and accuracy. Moreover, no bumping phenomenon has been found.
