妊娠期高血压大鼠血浆中GDF-15表达水平的测定 被引量:1

摘要 目的 :利用子宫胎盘缺血术诱发大鼠妊娠期高血压,测定妊娠期高血压大鼠血浆中生长分化因子(growth differentiafion factor-15,GDF-15)表达水平。方法:清洁级健康雌性Spragu-Dawley大鼠20只,随机分成2组,每组10只,分为假手术对照组及子宫胎盘灌注下降(reduced uterine perfusion pressure,RUPP)手术组。所有大鼠自然受孕,在妊娠期14 d对妊娠大鼠进行手术,在妊娠期13 d以及19 d分别测定大鼠血压。于妊娠19 d采血测定大鼠血浆GDF-15含量。结果:RUPP手术组妊娠19 d收缩压、舒张压与妊娠13 d相比均显著升高[(157.8±9.0)mmHg vs(118.3±5.7)mmHg;(122.5±6.9)mmHg vs(88.4±8.0)mmHg,P<0.05],假手术组则升高不明显。妊娠19 d RUPP手术组与假手术组相比,收缩压、舒张压均显著升高[(157.8±9.0)mmHg vs(117.0±9.4)mmHg;(122.5±6.9)mmHg vs(100.7±4.4)mmHg,P<0.05]。与假手术组相比,RUPP手术组妊娠19 d血浆中GDF-15浓度明显增高[(363.3±78.1)ng/L vs(276.7±69.4)ng/L,P<0.05)。结论:妊娠期高血压大鼠动物模型血浆中GDF-15表达水平明显增高,GDF-15是一个预测妊娠期高血压患者预后的潜在指标。 Objective:The aim of the study was to evaluate the growth differentiation factor-15(GDF-15) level in the reduced uterine perfusion pressure(RUPP) rat model by using uterine placenta ischemia to induce gestational hypertension in rats. Methods:A total of20 Spragu-Dawley rats were randomly divided into two groups with each group of 10 rats:the sham group and the RUPP group. The operation was taken on the 14 thday after gestation. On the 13 thand 19thday gestation,the blood pressure was measured. The blood was collected on the 19 thday gestation for the GDF-15 test. Results:In the RUPP rats,both the systolic pressure and diastolic pressure increased significantly on the 19 thday gestation compared with that on the 13 thgestation [(157.8 ± 9.0)mmHg vs(118.3 ± 5.7)mmHg,(122.5 ± 6.9)mmHg vs(88.4 ± 8.0)mmHg,P 〈 0.05],and the sham group had no significant changes. Compared with the sham group on19 thdays gestation, RUPP group rats were characterized by higher systolic pressure[(157.8 ± 9.0)mmHg vs(117.0 ± 9.4)mmHg;P 〈 0.05],higher diastolic pressure [(122.5 ± 6.9)mmHg vs(100.7 ± 4.4)mmHg;P 〈 0.05]. In the RUPP group,the GDF-15 level was significantly higher than that in the sham group [(363.3 ± 78.1)ng / L vs(276.7 ± 69.4)ng / L;P 〈 0.05]. Conclusion:GDF-15 level increased significantly in the gestation hypertension rats and is a potential cardiovascular prognostic maker for the gestation hypertension patients.
