优选维药复方益心颗粒挥发油的提取及β-环糊精包合工艺 被引量:3

摘要 目的优选维药复方益心颗粒中挥发油的提取及β-环糊精(β-CD)包合工艺。方法采用水蒸气蒸馏法提取挥发油,通过正交实验设计,以挥发油提取量为指标,优选挥发油的提取工艺;采用饱和水溶液法制备挥发油β-CD包合物,以包合物收得率和挥发油包合率为综合评价指标,通过正交实验考察挥发油与β-CD的比例、包合温度、包合时间对包合工艺的影响。结果最优提取工艺为:加10倍量水,浸泡8h,蒸馏5h;挥发油最佳包合工艺为:油与β-CD比例1∶10,包合温度40℃,包合时间2h。挥发油β-CD包合物具有一定的光稳定性、热稳定性、湿稳定性。结论优选的挥发油提取与包合工艺条件稳定可行。 Objective To optimize the extraction conditions and β-cyclodextrin(β-CD) inclusion process for volatile oil from Com-pound Yixin Granules .Methods Volatile oil was extracted by steam distillation method .The extracting technology was optimized by using the orthogonal design ,and the yield of volatile oil was used to evaluate the extraction process ;the β-CD inclusion complex of volatile oil was prepared by saturated aqueous solution method ,and the orthogonal test was used to optimize the inclusion tech-nology with ratio of volatile oil to β-CD ,inclusion time and temperature as factors ,and inclusion yield and volatile oil inclusion rate as indexes .Results The optimized volatile oil extraction conditions were to add 10 folds water into the mixture of crude drugs ,and to extract the herbal medicine for 5 h with previous soaking for 8 h with water .The optimized inclusion procedure was as follows :the proportion of volatile oil and β-CD was 1 ∶ 10 and stirring for 2 h at 40 ℃ .The inclusion complex of volatile oil-β-CD was stable to light ,heat and humidity .Conclusion The optimized extraction and inclusion conditions were reasonable and feasible .
