Expression of snail and slug in laryngeal squamous carcinoma and its relation to epithelial-mesenchymal transition,tumor invasion and metastasis

摘要 目的研究Snail、Slug基因在喉鳞状细胞癌中的表达,探讨其与肿瘤上皮-间质转化及侵袭转移之间的关系及临床意义。方法采用免疫组化EnVision法检测74例喉鳞状细胞癌组织及30例癌旁组织中Snail、Slug及Vimentin、E-cadherin的表达情况,统计分析其表达的相关性及与各临床病理指标之间的关系。结果 Snail、Slug蛋白在喉鳞状细胞癌组织中的阳性率为58.2%和55.4%,高于癌旁组织中阳性率(13.3%和16.7%,P<0.05)。Snail、Slug蛋白表达与E-cadherin呈负相关性(r=-0.567,P<0.001及r=-0.457,P<0.001),与Vimentin的表达呈正相关性(r=0.453,P<0.001及r=0.376,P<0.001)。Snai、Slug蛋白表达与分化程度、TNM分期及淋巴结转移密切相关(均P<0.05)。结论转录因子Snail、Slug可能通过参与与调控EMT过程,与喉鳞状细胞癌的发展和侵袭相关。 Objective To investigate the expression of snail and slug in laryngeal squamous carcinoma and its relation to epithelial- mesenchymal transition(EMT), tumor invasion and metastasis. Methods The expression of snail, slug, E- cadherin and vimentin was detected by immunihistochemistry in 74 specimens of laryngeal squamous carcinoma and 30 specimens of peri-cancerous mucosal tissue. Results The positive rates of snail and slug expression in laryngeal squamous carcinoma were 58.2%and 55.4%respectively, which were significantly higher than those in pericancerous mucosal tissue(13.3%and 16.7%, P〈0.05). The expression of snail and slug were negatively correlated with E- cadherin (r=- 0.567, P〈0.001 and r=- 0.457, P〈0.001) and positively corrected with vimentin (r=0.453, P〈0.001 and r=0.376, P〈0.001) in laryngeal squamous carcinoma. The expres-sion of snail and slug was closely correlated with pathological grade, TNM stage, thyroid cartilage involvement and lymph node metastasis of laryngeal squamous carcinoma. Conclusion Snail and slug are highly expressed in squamous laryngeal carcino-ma;it may be involved in epithelial- mesenchymal transition, invasion and metastasis of the tumor.
