摘要
[目的]研究长链非编码RNAs(lncRNAs)在刀豆蛋白A(ConA)诱导的小鼠免疫性肝损伤肝组织中表达谱的变化。[方法]利用lncRNAs芯片技术分别检测正常对照组小鼠及ConA诱导的免疫性肝损伤小鼠肝组织中lncRNAs表达谱,经对原始数据进行预处理达到均一化后,筛选出差异表达的lncRNAs并进行分析。[结果]与正常对照组小鼠相比较,ConA诱导的免疫性肝损伤在小鼠肝组织中表达1.5倍以上并有显著差异(P<0.05)的lncRNAs,确定为差异表达的lncRNAs。1.5倍以上变化的共60条;1.5倍以上升高的共17条;1.5倍以上降低的共43条;2倍以上升高共4条;2倍以上降低的共8条;3倍以上升高共2条;3倍以上降低的共2条。[结论]与正常对照组小鼠肝组织比较,ConA诱导的免疫性肝损伤小鼠肝组织中lncRNAs表达谱发生显著变化,提示差异性表达的lncRNAs可能参与了ConA诱导的小鼠免疫性肝损伤发生。
[Objective]To analyze the expression profile variation of long non coding RNAs(lncRNAs)in normal control and mice with immunological liver injury induced by ConA.[Methods]Use lncRNAs microarray technology to inspect the difference of lncRNAs expression profile between two groups,screen out the difference of lncRNAs after pretreatment and homogenization the raw data.[Results]Compared with normal control,60 lncRNAs have more than 1.5 times variation(P<0.05),while 17 increase more than 1.5 times and 43 reduce more than 1.5 times;4 increase more than 2 times and 8 reduce more than 2 times; 2 increase more than 3 times and 2 reduce more than 3 times.[Conclusion]Compared with the normal control group,lncRNAs expression profile change significantly in mice with immunological liver injury induced by ConA.Differential expression of lncRNAs may be involved in the development of immunological liver injury.
