摘要
目的 优化茯苓三萜类成分高效液相色谱指纹图谱分析方法,建立茯苓三萜类成分的指纹图谱.方法 色谱柱为Kromasil 100-5 C18柱;流动相为乙腈(A)-0.1%(体积分数,下同)H3PO4(B)溶液,梯度洗脱;检测波长为242 nm;柱温为30℃;流速为1.0 mL·min^-1.结果 不同来源的13批茯苓样品均标示出19个三萜类成分共有峰;采用中药色谱指纹图谱相似度评价软件(2004A版)进行相似度评价,13批样品的相似度为0.925~0.997,提示茯苓三萜类成分质量较稳定.结论 本方法准确可靠、重复性好,为茯苓质量控制提供了依据.
Objective To develop an HPLC method for establishment of fingerprint analysis on triterpenoids from Poria cocos.Methods HPLC was performed with the Kromasil 100-5 C18,and the acetonitrile(A)-0.1% phosphorous acid (B) solution (gradient elution) was employed with a flow rate of 1.0 mL · min^-1.The detection wavelength was 242 nm,and the column temperature was 30 ℃.Results 19 common peaks were separated from 13 batches of Poria cocos.The similarity was evaluated on the similarity evaluation system for chromatographic fingerprint of Traditional Chinese Medicine (2004 A edition).The similarities of 13 batches were 0.925-0.997,which indicated the quality of triterpenoids from Poria cocos was relatively stable.Conclusion The method is accurate and reproducible,which could be used for the quality control of Poria cocos.
