摘要
以391只藏绵羊为研究对象,采用PCR-SSCP技术分析孕酮受体(PGR)基因第4外显子在藏绵羊中单核苷酸多态性(SNPs),探讨PGR的生理功能及其对藏绵羊繁殖力的影响。结果表明:PGR基因扩增片段在藏绵羊品种中存在PCR-SSCP多态性,序列比对分析发现2个SNPs位点,全为颠换。在该群体中发现3种基因型,分别为AA、BB和AB,其中AA为优势基因型,A为优势等位基因。χ2独立性检验表明,PGR基因扩增片段上各基因型呈极显著差异。
This study was to analyze polymorphism of progesterone receptor (PGR) gene exon 4, which would provide the foundation basic for exploring the physiological functions of PGR and its influence to fecundity, and to clarify the plat eau breeding physiology mechanism of Tibetan sheep. 391 Tibetan sheep were used as experimental samples and SNPs of PGR gene exon 4 in Tibetan sheep were detected by PCR SSCP. The results showed that the polymorphism of exon 4 of PGR gene in Tibetan sheep was detected in this experiment. There were three genotypes named AA, AB and 13B in Ti betan sheep, AA was preponderant genotype, allele A was preponderant allele. The result of x^2 independence test indica- ted that the distribution and constitution of genotypes at the exon 4 of PGR gene showed significant differences in Tibetan sheep (P〈0.01).
