响应面分析法优化超临界CO_2萃取巨尾桉挥发油的工艺研究 被引量:2

Optimization of supercritical CO_2 extraction conditions for essential oil from Eucalyptus.grandis×Eucalyptus.urophylla using box-behnken design-response surface methodology

摘要 利用响应面法分析得到超临界CO2萃取巨尾桉叶挥发油的优化工艺条件为:萃取温度80℃,萃取时间8h,萃取压力40 MPa.在此条件下,巨尾桉叶挥发油的实际提取率可达7.86%.用气相色谱-质谱法对化学成分进行分析,以归一化法计算各个化学成分的相对含量.结果共分离出42个峰,鉴定了其中35个化学成分,占挥发油总量的98.31%.挥发油的主要成分中α~蒎烯占24.78%,桉油醇占45.57%,α~松油醇占2.51%,乙酸松油酯占8.34%. The Supercritical CO2 extraction process of essential oil from E. grandis × E. urophylla leaf was researched. The influences of three key factors were evaluated by response surface methodology. The results indicated that the optimal extraction conditions were as follows, extraction temperature of 80℃, extraction time of 8h, and extraction pressure of 40 MPa. Under the optimal extraction condi- tions, the yield of essential oil was 7.86%. The chemical constituents of the essential oil were analyzed by GC-MS and the relative contents of these constituents were calculated using square peaks to normali- zation. 42 peaks were separated and 35 constituents were identified, which were composed of about 98. 31% of the total essential oil. ConclusionThe main constituents are α~Pinene(24.78%), Cineole(45. 57%), alpha-Terpineol(2.51%), Terpinyl acetate(8.34%).
