摘要
目的探讨T-bet和GATA-3基因在肝泡型包虫病患者肝脏组织中的表达及作用。方法选取14例在新疆医科大学第一附属医院接受手术治疗的肝泡型包虫病患者病灶组织(L组)、病灶旁组织(P组)及正常肝组织(N组),采用实时荧光定量PCR(qRT-PCR)法检测各肝脏组织中T-bet和GATA-3 mRNA的表达,采用两两比较的t检验和Pearson相关性检验分析结果。结果 T-bet mRNA在L组、P组和N组的表达量分别为3.98、0.89和0.27,其中L组与P组比较差异无统计学意义(t=0.307,P〉0.05),L组与N组比较差异有统计学意义(t=3.695,P〈0.01),P组与N组比较差异有统计学意义(t=3.492,P〉0.05)。GATA-3mRNA在L组、P组和N组的表达量分别为119.23、6.30和0.53,其中L组与N组比较差异有统计学意义(t=5.190,P〈0.01),P组与N组比较差异有统计学意义(t=4.649,P〈0.01),L组与P组差异无统计学意义(t=1.711,P〉0.05)。肝脏不同组织中,T-bet和GATA-3mRNA的表达量呈正相关(相关系数为0.730-0.932)。结论 Th1细胞转录因子T-bet基因和Th2细胞转录因子GATA-3基因在肝脏病灶组织中的表达升高,这些变化或与泡型包虫病发病过程中寄生虫肉芽肿的形成有关。
Objective To investigate the expression and action of T-bet and GATA-3in liver tissue from patients with alveolar echinococcosis(AE). Methods Subjects were 14 patients with hepatic AE who were treated at the First Affiliated Hospital of Xinjiang Medical University.Liver tissue from these patients was divided into tissue from lesions(L),tissue around lesions(P),and normal liver tissue(N).mRNA was extracted from this tissue and then reverse-transcribed and amplified to yield cDNA.The concentration of T-bet and GATA-3was measured using real-time PCR(qRTPCR).A Student's t test and Pearson correlation were used for statistical analysis. Results The level of T-bet mRNA expression was 3.98 in group L,0.89 in group P,and 0.27 in group N.The level of expression was significantly higher in group L than in group N(t=3.695,P〈0.01).There was no significant differences in the level of expression in group L and group P(t=0.307,P〉0.05).There were significant differences in the level of expression in group P and group N(t=3.492,P〈0.01).The level of GATA-3mRNA expression was 119.23 in group L,6.30 in group P,and 0.53 in group N.The level of expression was significantly higher in group L than in group N(t=5.190,P〈0.01).There were no significant differences in the level of expression in group L and group P(t=1.711,P〉0.05).There were significant differences in the level of expression in group P and group N(t=4.649,P〈0.01).Expression of GATA-3and T-bet mRNA were correlated in different liver tissues. Conclusion Expression of the Th1 transcription factor T-bet and the Th2 transcription factor GATA-3increased in hepatic lesions.These factors may be involved in the process of parasitic granuloma formation.
