甲状腺功能亢进对大鼠红细胞系列指标及血清铁的影响

The impact of hyperthyroidism on erythrocyte indices and serum iron of rats

目的研究甲状腺功能亢进（简称甲亢）对大鼠红细胞系列指标及血清铁的影响。方法 SD雌性大鼠分为甲亢模型组和对照组,优甲乐灌胃制造甲亢模型,对照组灌服生理盐水。分别在灌胃给药后的第1~8周采血,采用放射免疫法测定大鼠血清三碘甲状腺原氨酸（T3）、四碘甲状腺原氨酸（T4）及促甲状腺激素（TSH）含量;使用全自动血细胞分析仪测定大鼠红细胞系列指标;比色法测定血清铁含量和总铁结合力。结果甲亢模型组大鼠血清T3、T4含量增高,而TSH含量下降。甲亢模型组大鼠红细胞计数（RBC）在第2~8周均明显高于对照组（P〈0.05）;红细胞压积（HCT）在第1~8周均明显高于对照组（P〈0.01）;平均红细胞体积（MCV）在第2~8周均明显低于对照组（P〈0.05）;平均细胞血红蛋白浓度（MCHC）在第1~8周均明显低于对照组（P〈0.01）;红细胞体积分布宽度标准差（RDW-SD）和红细胞体积分布宽度变异系数（RDW-CV）在第1~2周均高于对照组（P〈0.01）;血清铁含量和总铁结合力在第4~8周均高于对照组（P〈0.01）。结论本实验甲亢模型鼠虽未出现明显的贫血表现,但红细胞有明显的低色素性改变,血清铁含量和血清铁结合力均升高,提示有铁利用障碍。

Objective To study the change of erythrocyte indices and serum iron in hyperthyroidism rats.Methods SD female rats were randomly divided into hyperthyroidism group and control group.Hyperthyroidism rats were induced by intragastric administration of Euthyrox（Levothyroxine）and their blood and serum were obtained after administration of 1-8weeks,respectively.And the control rats were given normal saline.Levels of T3,T4 and TSH were determined by a radioimmunoassay.Erythrocyte index were measured by automatic blood cell analyzer.Serum iron concentration and total iron binding capacity were determined by chromatometry.Results The concentration of serum T3 and T4increased in hyperthyroidism rats,whereas the TSH decreased.RBC was higher in hyperthyroidism rats at 2-8weeks compared with the control rats（P〈0.05）.HCT was higher in hyperthyroidism rats at 1-8weeks compared with the control rats（P〈0.01）.MCV was lower in hyperthyroidism rats at 2-8weeks compared with the control rats（P〈0.05）.MCHC was lower in hyperthyroidism rats at 1-8weeks compared with the control rats（P〈0.01）.RDW SD and RDW CV was higher hyperthyroidism rats at 1-2weeks compared with the control rats（P〈0.01）.Serum iron concentration and total iron binding capacity increased in hyperthyroidism rats at 4-8weeks compared with the control rats（P〈0.01）.Conclusion The hyperthyroidism rats in the present study is absent significant manifestation of anemia,whereas the hypochromic changes of erythrocyte is significant.Serum iron concentration and total iron binding capacity increase in hyperthyroidism rats,which demonstrate the iron utilization disorder.
