摘要
为了使重组GST-Apoptin融合蛋白能够与肿瘤细胞靶向性结合,并发挥其特异性诱导肿瘤细胞凋亡的作用,本文采用化学方法将叶酸连接到GST-Apoptin融合蛋白,实验结果表明叶酸与GST-Apoptin融合蛋白的偶联率为6%,最佳偶联条件是20 mg/m L活化叶酸在p H 10.0条件下室温反应60 min。叶酸化的GST-Apoptin融合蛋白(folate-GST-Apoptin)具有显著诱导肿瘤细胞凋亡作用,1.0μg/m L的folate-GST-Apoptin可使50%的肿瘤细胞(KG-1)凋亡,而对正常人外周血淋巴细胞没有诱导凋亡作用。
To integrate fusion protein of recombined GST-Apoptin with cancer cell targeting so as to exert its func- tion of peculiar induced tumor cell apoptosis.The paper adopts chemical process to connect folic acid with fusion pro- tein of GST-Apoptin, and experiments show that the coupling rate of folic acid and fusion protein of GST-Apoptin is 6%, and the best coupling condition is 20 mg/mL excited folic acid reacts for 60 minutes in indoor temperature with pH 10.0.Folate-GST-Apoptin bears remarkable function of induced tumor cell apoptosis,and 1.0 Ixg/mL fo- late-GST-Apoptin can achieve 50% of tumor cell (KG-1)apoptosis,while having no function of induced tumor cell apoptosis on peripheral blood lymphocyte of normal people.
