IL-17A在肺部铜绿假单胞菌感染小鼠中的表达及其意义

摘要 目的:探讨IL-17A在肺部铜绿假单胞菌感染小鼠中的表达及其作用。方法:30只SPF级昆明白雌性小鼠,随机分成对照组和模型组,模型组小鼠气管内滴入铜绿假单胞菌悬液,对照组小鼠气管内滴入生理盐水,造模后第3、9、24 h通过心脏采血处死小鼠,每次各5只。将小鼠的肺组织制做成病理切片,HE染色观察肺组织炎症变化,免疫组化法检测肺组织IL-17A的表达,ELISA法检测小鼠血清中IL-17A的浓度。结果:1与对照组相比,随着时间变化,模型组肺泡结构破坏逐渐加重,肺泡腔炎症细胞浸润逐渐明显。2与对照组相比,模型组肺组织IL-17A各个时间点的表达明显增加,随着时间的延长而增加(P<0.05)。3与对照组相比,模型组血清中IL-17A的浓度在造模后3 h无明显差异,造模后9 h和24 h明显升高,差异有统计学意义。4血清中IL-17A与肺泡炎积分呈正相关(r=0.852,P=0.00)。结论:IL-17A可能通过炎症细胞的募集参与了铜绿假单胞菌肺部感染的炎症反应及早期对病原菌的清除过程。 Objective: To investigate the expression and role of IL-17 A in lungs of mice infected with pseudomonas aeruginosa. Methods:Thirty SPF Kunming white female mice were randomly divided into control group and model group. Model group was instilled intratrachea with 0.3ml pseudomonas aeruginosa suspension,while control group with the same volume of normal saline. The mice were killed by intracardiac sampling blood3, 9, 24 h after the model was built,5 mice each time. Lung tissue in mice was taken to prepare histopathologic slide for the observation of inflammatory change in lung through HE staining, and expression of IL-17 A in lung through immunohistochemistry as well as expression of IL-17 A in serum by ELISA. Results: In the model group,the destruction of alveolar structure and the infiltration of inflammatory cells were increasing with the time; the expression of IL- 17 A in the lung tissue was raised at each time point, the difference was statistically significant;the expression of IL- 17 A in mice serum began increased 9 hours after the model were built and more significant at 24 h, the difference was statistically significant. Serum IL-17 A and scores of alveolitis were positively correlated. Conclusion: IL-17 A might be involved in the inflammatory response and early elimination of pathogen pseudomonas aeruginosa through the recruitment of inflammatory cells.
