摘要
目的:观察醒脑开窍针刺法对脑缺血再灌注损伤大鼠增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达及神经功能的影响。方法:将20只大鼠随机分为假手术组、模型组、醒脑开窍针刺法治疗组,剔除死亡大鼠后每组各5只。假手术组只分离颈内外动脉,其余两组用线栓法建立大脑中动脉缺血再灌注(middle cerebral artery occlusion/reperfusion,MCAO/R)损伤模型。用Longa评分法评估大鼠神经行为学障碍程度,用免疫组化染色法观察缺血侧额叶皮层区以及侧脑室室管膜下区(subventricular zone,SVZ)PCNA的阳性表达。结果 :再灌72 h后,与模型组比较,针刺组神经行为学评分明显降低(P<0.05),缺血侧SVZ及皮层区PCNA的阳性表达显著增加(P<0.01,P<0.05)。结论:醒脑开窍针刺法可能通过提高缺血再灌注损伤大鼠PCNA的表达,促进神经细胞的增殖和DNA的修复从而改善其神经行为功能。
Objective: To explore the effect of xingnao kaiqiao acupuncture on the expression of PCNA and neurological function in rats with cerebral ischemia-reperfusion injury. Methods: 20 rats were randomly divided into 3 groups: sham operation, model and xingnao kaiqiao acupuncture treatment group.In sham operation group,only internal and external carotid artery were isolated. The model of middle cerebral artery occlusion/reperfusion(MCAO/R) was established by suture embolus method. After surgeries 15 rats survived, 5 each group.The neurobehavioral impairment was assessed by Longa method, the expression of PCNA was observed in the cortex and SVZ of lateral ventricle by immunohistochemical staining. Results: After 72 h of reperfusion, the neurological deficit scores significantly decreased(P 〈0.05)and the expression of PCNA in SVZ and cortex was significantly increased in acupuncture treatment group compared with model group(P〈0.01; P〈0.05). Conclusion: Xingnao kaiqiao acupuncture can improve the impaired neurobehavioral function in rats with ischemia-reperfusion injury by increasing the expression of PCNA and repairing of injured DNA of nerve cells.
