摘要
以辣椒雄性不育系为外植体,研究分析辣椒植株再生过程中经常遇到的外植体褐化问题,找出辣椒组培过程中污染率最低的外植体消毒处理方法,并对影响辣椒组培中外植体褐化程度的因素进行探索。结果表明:较低的pH值(5.5)可降低辣椒不育系外植体褐化率,更有利于愈伤的生长;PPO、POD、CAT、SOD活性及MDA含量在培养基中添加1.0 mg/L6-BA时比添加相同浓度的2,4-D时要高;培养基中添加的三种附加物按防褐化效果的大小依次为PVP、Vc、AC。
The pepper male sterile line was used as experimental materials.During pepper regeneration,one of the diffcult issues was browning.This study found out the best method of disinfection and explored the factors affecting the degrees of browning of pepper male sterile line.The results showed that relative low pH value (5.5) brought down browning mtio of pepper male sterile line,hence callus grew much better.The enzymatic activity of PPO,POD,CAT,SOD and the content of MDA were higher with additional 1.0mg/L 6-BA into the culture medium than adding the same concentration of 2,4-D into the cuhure medium.The preventable effects of browning of the three different kinds of appendage were arranged in descending as PVP,Vc and AC.
