笹之雪组织培养和植株再生体系研究

摘要 通过对笹之雪(Agave victoriaereginae)不同外植体、不同激素配比的研究,建立笹之雪离体培养高效植株再生体系。笹之雪侧芽在含2.0 mg·L-16-BA和0.2 mg·L-1NAA的MS培养基中增殖效果较好,增殖系数4.6。比较分析叶片各部位愈伤组织诱导和再生能力,结果表明:叶基>叶中>叶顶,且只有叶基诱导出有效愈伤组织,是较好的外植体材料;叶片各部位在2.0 mg·L-16-BA、1.0 mg·L-1KT和0.2 mg·L-1NAA的MS培养基上愈伤组织的诱导率达到最高,叶基、叶中和叶顶诱导率分别为100%、98%、89%,大部分叶基还能通过愈伤组织间接分化形成不定芽。将有效愈伤组织转至分化培养基上,在MS+6-BA 1.0 mg·L-1+KT 1.5 mg·L-1+NAA 0.2 mg·L-1培养基上的总分化芽数最多,长势也较其他配方的好。MS+6-BA 0.5 mg·L-1+KT 1.0 mg·L-1+NAA 0.2 mg·L-1对不定芽的增殖效果最好,增殖率100%,平均增殖系数5.5。在不定芽生根中,以MS+KT 1.5 mg·L-1+NAA 0.5 mg·L-1效果较好,出根率高,根系粗壮。 Different Agave victoriaereginae explants and the compounding proportions of different hormones were comparatively studied in tissue culture and thereafter a high-efficient plant-regeneration system by Agate victoriaereginaein vitro culture was established.The medium MS + 6-BA 2.0 mg· L-1 + NAA 0.2 mg · L-1 was the best to multiply shoot from lateral bud of Agate victoriaereginae,with multiplication coefficient of 4.6.The sequence of callus induction and regeneration of different places in 1eaf was leaf base > leaf mid > leaf top.Effective callus was ouly induced from leaf base and they were great explants materials.The induction medium MS + 6-BA 2.0mg · L-1 + 1.0 KT mg · L-1 + NAA 0.2 mg · L-1 was the best to induce callus from different places in 1eaf and leaf base had 100 % regeneration rate,leaf mid 98 % and leaf top 89 %.And most leaf base could indirectly divide small shoot from callus.Effective callus transferred to the division medium.The results showed that the medium MS + 6-BA 1.0 mg· L-1 + KT 1.5 mg+ NAA 0.2 mg · L-1 produced most differentiation shoots and the growth trend in it was also better than in others.The multiplication effect of shoot was the best in MS + 6-BA 0.5 mg · L-1 + KT 1.0 mg · L-1 + NAA 0.2 mg· L-1,with multiplication rate of 100 % and the average multiplication coefficient of 5.5.In adventitious shoot regenerating,MS + KT 1.5 mg · L-1 + NAA0.5 mg · L-1 was the beet formulation,with high rooting frequency and strong root system.
