含大承气汤动物血浆对三磷酸肌醇间质细胞Cajal受体表达的影响

Influence of animal plasma with Dachengqi decoction on the expression of triphosphoinositide interstitial cells Cajal receptor in rats

摘要 目的:探讨含大承气汤动物血浆对三磷酸肌醇间质细胞Cajal受体表达的影响。方法:将60只断乳的12周龄大鼠构建Wistar大鼠模型。将所有的大鼠按照随机数字表法分为正常大鼠血浆组,高浓度大鼠血浆组,低浓度大鼠血浆组。不同剂量的大承气汤动物血浆作用于Wistar大鼠间质细胞Cajal受体(ICC),利用逆转录酶PCR(RT-PCR技术)检测不同组中三磷酸肌醇受体IP3R以及Ryanodine受体(RyR受体)的表达。结果:IP3R基因有3种类型分别为IP3R1、IP3R2、IP3R3与β-actin的PCR相对量,其中高浓度大鼠血清组,低浓度大鼠血清组的相对量与正常大鼠血清组相对量明显的升高,高浓度大鼠组升高的更明显,3组经过方差分析差异具有统计学意义(P<0.05),两两比较高浓度大鼠组与正常组以及低浓度大鼠组与正常组之间的差异具有统计学意义(P<0.05);RYR基因有3种类型分别为RYR.1、RYR.2、RYR.3的PCR相对量,3组经过方差分析差异具有统计学意义(P<0.05),两两比较高浓度大鼠组与正常组以及低浓度大鼠组与正常组之间的差异具有统计学意义(P<0.05)。结论:大承气汤动物含药血清可显著增强大鼠小肠三磷酸肌醇间质细胞Cajal受体IP3R表达以及RYR基因,并存在量效依赖关系。 OBJECTIVE To explore the influence of animal plasma with Dachengqi decoction on the expression of triphosphoinositide interstitial cells Cajal receptor in rats.METHODS 12 weeks old,60 rats were weaned to construct the model of Wistar rats.All the rats were randomly divided into the group of normal rat plasma,high concentration Dachengqi decoction in rat plasma group,low concentration in rat plasma group.The effects of different doses of Dachengqi decoction animal plasma on Wistar rats' interstitial cells Cajal receptors(ICC)were studied.,using reverse transcriptase PCR(RT-PCR)to detect the expression of triphosphoinositide IP3 R receptor and ryanodine receptor(RyR receptor)of three different groups.RESULTSIP3 R gene was the relative amounts of three types IP3R1,IP3R2,IP3R3 and beta-actin PCR,the relative amounts in the high concentration Dachengqi decoction of rat serum group and in the low concentration of rat serum group were significantly elevated as compared with normal serum group rats,and in the high concentration group increased more significantly,there was statistically significant difference after the variance analysis of the difference(P〈0.05).There was significant difference between the high concentration of rats group with the normal group,and significant difference between the low concentration of rats group with the normal group(P〈0.05);RYR gene was the relative amount of three types RYR.1,RYR.2,RYR.3PCR,there were significant differences between the high concentration of rats group with the normal group,and the differences between the low concentration of rats group with the normal group(P〈0.05).CONCLUSION Dachengqi decoction animal serum can significantly enhance the expression of triphosphoinositide IP3 R receptor and ryanodine receptor(RyR receptor)gene of interstitial cells Cajal receptor in rats,and existing dose dependent relationship.
