人参皂苷Rb1对抑郁症大鼠海马5-HT及5-HT1A受体表达的影响 被引量:13

Influence of Ginsenoside Rb1 on 5-HT Level and Expression of 5-HT1A Receptor in Hippocampus of Depressive Rats

摘要 目的研究人参皂苷Rb1对抑郁大鼠海马5-HT及5-HT1A受体的作用,探讨人参皂苷Rb1抗抑郁作用机制。方法选用成年Wistar大鼠45只,分为对照组、模型组和治疗组。给予慢性不可预性温和应激(CUMS)建立抑郁模型,治疗组给予人参皂苷Rb1。采用糖水偏好实验、高架十字迷宫实验和Morris水迷宫实验进行行为学检测,采用高效液相色谱法测定海马5-HT浓度,Western blot检测5-HT1A受体表达。结果模型组大鼠糖水偏好百分比明显降低(P<0.05),进入开臂次数和停留时间百分比增高(P<0.05),水迷宫逃避潜伏期延长(P<0.05);治疗组能逆转这些改变;模型组大鼠海马5-HT含量和5-HT1A受体表达水平明显低于对照组(P<0.05),治疗组5-HT含量和5-HT1A受体表达水平明显升高,但仍低于对照组(P<0.05)。结论人参皂苷Rb1抗抑郁作用可能与影响海马5-HT水平及5-HT1A受体表达有关。 Objective To observe the effect of ginsenoside Rb1 on 5-HT level and expression of 5-HT1 A receptor in hippocampus of depressive rats. Methods Forty-five male Wistar rats were randomly divided into control group,model group and treatment group. The depression animal model was produced by giving the rats chronic unpredicted mild stress. The depressional behavior was examined using sucrose preference test,Elevated Plus Maze test and Morris water maze test. The level of 5-HT and expression of 5-HT1 A receptor protein were detected using high performance liquid chromatography(HPLC) and Western blot respectively. Results Change in percentage of sucrose of model group and treatment group rats were significantly lower,while the percentage of the number of entry into open arms and percentage of time spent were higher,than those of control group rat(P〈0.05). These indexes were reversed in treatment group. The content of 5-HT and expression of 5-HT1 A receptor protein of model group and treatment group were higher than that of control group,while the index was lower after treatment with bupleurum liver-coursing powder(P〈0. 05). Conclusion Effect of ginsenoside Rb1 antidepressant may induced by affect the content of 5-HT and expression of 5-HT1 A receptor in hippocampus.
