摘要
目的利用高效液相色谱-蒸发光散射检测法测定湖北贝母中湖贝甲素、贝母素乙和贝母素甲的含量。方法 Agilent ZORBAX SB-C18(4.6mm×250mm,5μm)色谱柱,流动相:乙腈-0.02%二乙胺溶液(75∶25),流速1.0 ml·min^-1,检测器为Alltech 2000 ELSD,漂移管温度为88℃,载气流速为2.2L·min^-1,对数外标两点法定量。结果湖贝甲素在0.304-3.648μg范围内呈线性,回收率为100.25%,RSD为2.16%;贝母素乙在2.132-25.584μg范围内呈线性,回收率为97.65%,RSD为1.89%;贝母素甲在0.297-3.562μg范围内呈线性,回收率为96.82%,RSD为1.93%。结论测定方法分离好,重现性分离度均符合要求,可用于湖北贝母药材的质量监控。
Objective To determine the content of hupehenirine,peiminine and peimine in Bulbus Fritillariae Hupehensis by using a new HPLC-ELSD methods. Methods Agilent ZORBAX SB-C18column(4. 6mm × 250 mm,5μm) was used,the mobile phase was consisted of Acetonitrile-0. 2% diethylamine(75:25),at 1 m L·min^-1flow rate. Alltech 2000 ELSD detector was used,the drift tube temperature and nitrogen carrier gas flow rate of the ELSD were set at 88℃ and 2. 2L·min-1respectively.Quantitative was analysed by external standard method. Results Within 0. 304 - 3. 648 μg hupehenirine had a good linearity,the average recovery was 100. 25% with RSD 2. 16%; Within 2. 132 - 25. 584 μg peiminine had a good linearity,the average recovery was 97. 65% with RSD 1. 89%.; Within 0. 297 - 3. 562 μg peimine had a good linearity,the average recovery was 96. 82%with RSD 1. 93%. Conclusion The method is sensitive and reproducible and it could be used for quantitative determination of Bulbus Fritillariae Hupehensis.
