摘要
目的探讨邻苯二甲酸二(2-乙基己基)酯(DEHP)对雄性大鼠睾丸的病理损害和超微结构的影响。方法 5周龄雄性SD大鼠40只,随机分为对照组、DEHP低剂量组、DEHP中剂量组、DEHP高剂量组,每组10只,每天灌胃1次,每周5次,连续灌胃6周。于末次染毒24 h后处死动物,称量大鼠体重及睾丸湿重,并计算睾丸系数;苏木精-伊红染色(HE染色)后进行光镜和电镜检查。结果与对照组相比,DEHP中剂量组及高剂量组睾丸重量、睾丸脏器系数明显降低;病理检查发现,DEHP中剂量组及高剂量组睾丸支持细胞形成大量空泡,睾丸间质细胞异常增生,生精细胞严重脱落;电镜检查发现,DEHP中剂量组及高剂量组睾丸内线粒体断脊、脱颗粒,精细胞内的线粒体鞘异常,精原细胞内大量空泡。结论 DEHP可引起大鼠睾丸明显病理损害和超微机构改变,产生雄性生殖毒性。
Objective To explore the testicular pathological damage and ultrastructure change after di-2-ethylhexyl phthalate(DEHP) treatment in male rats.Methods A total of 40 healthy 5-week-old male Sprague-Dawley rats were randomly divided into four groups,namely control group,DEHP low dose group,DEHP medium dose groupand DEHP high dose group.Rats were exposed to DEHP via gavage administration once a day,five times a week,and DEHP administration lasted for six weeks.After DEHP treatment,the rats were anesthetized through ether,then sacrificed for the further study.The testis and testis coefficient were calculated,histopathological change of the testis was examined by HE(hematoxylin-eosin) staining and the ultrastructure of testis was examined by transmission election microscopy.Results Compared with control group,the wet weight of testis and testis coefficient were significantly decreased in DEHP medium dose group and DEHP high dose group.Pathological examination found that a large number of vacuoles appeared in sertoli cells in DEHP medium dose and high dose group,leydig cells showed abnormal hyperplasia and sperm cells loss seriously.Electron microscopy examination found that mitochondria form broken ridges and degranulation in DEHP medium and high dose group,a large number of vacuoles existed in spermatogonium and sertoli cells,sperm cells showed dysplasia.Conclusion DEHP could induce serious pathological damage and ultrastructure change in rat testis,then lead to male reproductive toxicity.
