Biological effects of low-intensity pulsed ultrasound on cells

摘要 目的研究低剂量脉冲超声波(low-intensity pulsed ultrasound,LIPUS)对体外培养细胞的生物学作用。方法以LIPUS作用于人皮肤成纤维细胞0~40 min,采用MTT比色法、酶谱法和Western blot法观察细胞增殖效应。以LIPUS作用于人皮肤成纤维细胞和人胰腺癌细胞(Mia Pa Ca-2)0~100 min,用Annexin V法检测凋亡效应。结果在0~40 min LIPUS刺激下,细胞活性、细胞所分泌的活性介质等增加,细胞内蛋白磷酸化水平升高;LIPUS刺激超过60 min后,凋亡细胞数增加,且LIPUS刺激达100 min时,凋亡细胞数增加更加明显。结论短时间LIPUS刺激能促进细胞增殖,超过一定时间则会抑制细胞生长,甚至引起细胞凋亡。 Objective To understand the biological effects of low-intensity pulsed ultrasound(LIPUS) on the cultured cell in vitro. Methods LIPUS was used to stimulate the human skin fibroblasts for 0-40 min, and the effects of cell proliferation were determined by MTT assay, metalloproteinase assay and Western blotting assay. LIPUS was used to stimulate the human skin fibroblasts and human pancreatic cancer cell strains(Mia Pa Ca-2) for 0-100 min, and the effects of cell apoptosis were determined by Annexin V assay. Results LIPUS treatment of 0-40 min significantly increased the cell proliferation, the activity of extracellular matrix metalloproteinases(MMPs) and the level of tyrosine phosphorylation. In addition, cell proliferation was inhibited and cell apoptosis increased when the stimulating time exceeds 60 min. The number of apoptotic cells increased more obviously by the stimulation of LIPUS for 100 min. Conclusion LIPUS can induce cell proliferation during comparative shorter time and cell apoptosis during longer time in vitro.
