摘要
目的建立同时测定茶碱和多索茶碱血清浓度的高效液相色谱法。方法血清样本经甲醇沉淀蛋白后直接进样分析,色谱柱为Waters C18(5μm,4.6mm×250mm)柱,流动相为甲醇-水(55∶45),检测波长为235nm。结果一定浓度范围内(茶碱3.8~60.0mg/L,多索茶碱2.5~50.0mg/L),2种药物的峰面积与浓度呈良好线性关系,回收率均高于95%(95%~104%,n=9),日内变异〈5%(n=9)。结论高效液相色谱法同时测定茶碱和多索茶碱的血清药物浓度操作简便,结果稳定可靠。
Objective To establish a performance liquid chromatography method for the determination of theophylline and doxofylline in human serum.Methods The proteins were precipitated by adding methanol in the serum samples,the chromatographic column was Waters C18(5μm,4.6mm×250mm)column,the mobile phase was a mixture of methanol and water(55∶45),and the detection wave length was 235 nm.Results In a certain range of concentration(theophylline:3.8to 60.0mg/L,doxofylline:2.5to 50.0mg/L),there was a good linear relationship between peak area and concentration of these two drugs,the recovery rates were higher than 95%(95%to 104%,n=9),and the within day variation was less than 5%(n=9).Conclusion The method is simple,stable and reliable.
