摘要
目的:研究低毫安(10m A)电化学治疗对人乳腺癌细胞阿霉素耐药株(MCF-7/ADR)多药耐药(MDR)性的逆转作用及其机制。方法:用MTT法测定电化学治疗对细胞的生长抑制作用,荧光分光光度法检测细胞内阿霉素(ADR)的浓度,流式细胞术检测细胞P-糖蛋白(P-gp)的表达。结果:低毫安电化学治疗(0.1C-2C)能不同程度降低ADR对MCF-7/ADR细胞的IC50;2C的电化学治疗能显著提高ADR在MCF-7/ADR细胞内的浓度,降低MCF-7/ADR细胞P-gp的表达。结论:电化学治疗能逆转MCF-7/ADR细胞的MDR,其机理可能是抑制了P-gp的功能表达,增加了细胞内ADR的积累。
Objective: To investigate the reversal effects of electrochemical therapy( ECT) on adriamycin resistant human breast cancer cell line( MCF-7 / ADR) and its mechanisms. Methods: The growth inhibition of ECT was determined by MTT assay. The intracellular adriamycin( ADR) accumulation was detected by fluorospectrophotometry assay. The expression of P-glycoprotein( P-gp) was examined by flow cytometric( FCM) assay. Results: ECT( 0. 5C-2C) could decrease the IC50 of ADR in MCF-7 / ADR cells. 2C ECT could significantly increase the intracellular concentration of ADR and reduce the expression of P-gp in MCF-7 / ADR cells. Conclusion: ECT can reverse multidrug resistance of MCF-7 / ADR cells. The reversal mechanisms were associated with the increase of the intracellular accumulation of ADR resulted from decrease of P-gp pump-efflux activity and the expression of P-gp in MCF-7 / ADR cells.
