摘要 目的:构建表达N-myc下游调节基因2(NDRG2)两种亚型(长短2种亚型分别命名为NDRG2L、NDRG2S)的重组逆转录病毒载体,观察逆转录病毒介导的NDRG2基因对卵巢癌HO-8910细胞增殖抑制的影响。方法:以携带人NDRG2基因的两种亚型逆转录病毒载体转染体外培养的卵巢癌细胞株HO-8910。采用Western blot检测目的基因的表达,MTT、平板克隆实验检测对细胞增殖能力的影响。结果:细胞经逆转录病毒转染后,Western blot检测NDRG2两亚型蛋白特异表达。MTT、平板克隆实验结果显示NDRG2基因两种亚型对HO-8910细胞生长均有明显促进作用(P<0.05),而二者之间无明显差别。结论:逆转录病毒载体使卵巢癌细胞表达NDRG2基因,可以明显促进HO-8910细胞的生长和增殖。 Objective: To construct p LNCX2- NDRG2,p LNCX2- NDRG2 S recombinant retroviral vector and to observe its effect on the proliferation of ovarian cancer HO- 8910 cells. Methods: The p LNCX2- NDRG2 L,p LNCX2- NDRG2 S were transfected into ovarian cancer HO- 8910 cells,and the protein expression of NDRG2 was detected by Western blot. The effect on the proliferation of cells were detected by MTT and plate cloning test. Results: The NDRG2 L,NDRG2S protein was efficiently expressed in ovarian cancer HO- 8910 cells. MTT and plate clone experimental results showed that the 2 isoforms of NDRG2 gene significantly promote the growth of the HO- 8910 cells( P 0. 05),but no significant difference between the two isoforms. Conclusion: The growth and proliferation of ovarian cancer HO- 8910 cells were significantly promoted by NDRG2 L and NDRG2S.
