摘要
目的:观察人食管鳞癌细胞及组织中miRNA-381的表达变化,探讨其与放射敏感性的关系。方法:采用RT-PCR法测定人食管鳞癌细胞TE-1、TE-10、TE-11及20例符合纳入标准的食管鳞癌组织中miRNA-381的表达。完成放疗后1个月接受影像学复查,分析患者的近期放疗疗效。采用克隆形成实验检测细胞的放疗敏感性。结果:入组患者根据近期疗效分为放疗抵抗组(6例)和放疗敏感组(14例),放疗抵抗组的miRNA-381表达显著低于放疗敏感组(P<0.05)。克隆形成实验结果显示细胞的放疗敏感性为TE-11>TE-10>TE-1,且有显著统计学差异(P<0.05)。RT-PCR结果显示放疗最敏感的TE-11细胞中miRNA-381表达量最高,而放疗抵抗的TE-1细胞中miRNA-381表达量最低。近期疗效和食管鳞癌组织中miRNA-381相对表达量的相关性分析显示miRNA-381相对表达量与近期疗效呈正相关(r=0.72,P<0.05)。结论:MiRNA-381在放疗敏感的食管鳞癌细胞及临床标本中的表达量显著高于放疗抵抗的细胞及组织。MiRNA-381相对表达量越高的食管鳞癌患者接受根治性放疗后的近期疗效越好。
Objective: To investigate the expression of miRNA-381 in esophageal squamous cell carcinoma( ESCC) cells and tissue,and its correlation with radiosensitivity. Methods: The expression of miRNA-381 in ESCC cell lines( TE-1,TE-10,and TE-11) and ESCC tissue were detected by quantitative real-time PCR( RT-PCR). The short-term effects of radical-radiotherapy in ESCC were analyzed by imageological examination and cell radiosensitivity was measured by colony formation assay. Results: ESCC patients met our inclusion criteria were divided into radioresistant group( n = 6) and radiosensitive group( n = 14) by the short-term effects. MiRNA-381 was higher expressed in radiosensitive group compared with the radioresistant group( P 0. 05). According to the radiosensitivity assay,exprssion of miRNA-381 in the most radiosensitive cell TE-11 was higher than that in the most radioresistant cell TE-1( P 0. 05). Correlation analysis showed that miRNA-381 expression in ESCC patients was positively correlated with the short-term effects after radical-radiotherapy( r = 0. 72,P 0. 05). Conclusion: Higher expression of miRNA-381 prognosticates radiosensitivity for both patients and cell lines,and higher miRNA-381 expression associated with better short-term effects of radical-radiotherapy treatment in ESCC.
