核纤层蛋白基因产物与衰老及老年性相关疾病关系研究进展

Research on the progress of relationships between LMNA products and aging or age-related diseases

摘要 核纤层是内核膜下约10 nm厚的蛋白质层,位于内层核膜与染色质之间,向外与内层核膜上的蛋白结合,向内与染色质的特定区段结合,构成贯穿于细胞核和细胞质统一网架结构体系;是核大小及形状的主要决定因素,并且参与DNA复制及转录等基本活动.核纤层主要结构成分是纤层蛋白(lamins),人类核纤层由A型、B型两种lamins构成,分别由Lamin A基因(LMNA)、Lamin B(LMNB)基因编码.lamins对维持细胞结构及功能发挥重要作用,LMNA基因产物在正常细胞生理性老化中可能也发挥作用.我们就LMNA基因产物及其加工过程,lamins与衰老的关系及机制,lamins对动脉粥样硬化、脂代谢紊乱、中枢神经系统疾病等老年性相关性疾病的影响作一综述.
