摘要
为降低蝴蝶兰组培苗生产成本,简化组培程序,以自主研发的抗菌剂进行蝴蝶兰类原球茎扩繁、继代和增殖苗壮苗生根的抗菌组培(开放组培)试验。结果表明:附加适当浓度抗菌剂能有效抑制培养基污染,类原球茎扩繁培养后增重倍数与对照(高压灭菌培养基)差异不显著。类原球茎在抗菌培养基继代培养2次(合计培养150 d),增重倍数与对照无明显差异。增殖苗在含香蕉汁培养基壮苗生根,2个适宜浓度抗菌剂的培养基污染率与对照基本相同;瓶苗成活率、叶片数、根数都与对照相同,平均苗高、平均根长均与对照无明显差异。所以该抗菌剂应用于蝴蝶兰的抗菌组培是可行的。
To reduce the production cost of tissue-cultured plantlets of Phalaenopsis and simplify the tissue culture process, the proliferation and subculture of Phalaenopsis protocorm-like bodies (PLBs) as well as the plantlet growth and rooting were studied by using the self developed antimicrobial agent. The results showed that a proper concentration of the antimicrobial agent could effectively inhibit the microbial contamination of culture media;After proliferation the weight gain multiples of PLBs were not significantly different from those of the control(on autoclave sterilized media) ,and after subculturing the PLBs two times(for 150 days in all),the weight gain multiples of PLBs were also not significantly different from those of the control;The proliferated plantlets grew and rooted on the media containing mashed banana,and the two kinds of media supplemented with a proper concentration of the antimicrobial agent had basically the same contamination rate as the control;The survival rates,leaf numbers and root numbers of plantlets in bottles were the same as those of the control,and the average plant height and average root length were also not significantly different from those of the control. From the above results it was concluded that Phalaenopsis tissue culture with antimicrobial is practical.
