蓝莓花色苷对放射性心脏损伤的保护机制研究 被引量:4

摘要 目的:探讨IL-1和TGF-β在放射所致心脏损伤组织中的表达变化及蓝莓花色苷干预的影响。方法:将40只大鼠随机分成空白对照组、单纯照射组、20mg/kg蓝莓花色苷照射组、80mg/kg蓝莓花色苷照射组,每组各10只。照射前蓝莓花色苷灌胃给药,然后通过单次全胸15Gy照射剂量进行照射,照射后每天对大鼠的状态进行观察并继续灌胃。饲养8d后,对大鼠心肌酶谱指标进行检测,苏木精-伊红及Masson染色观察心肌组织病理变化,运用荧光定量PCR仪对IL-1和TGF-β相对表达量的变化进行分析。结果:与空白对照组比较,单纯照射组的苏木精-伊红及Masson染色显示心肌有一定程度的损伤,血清谷草转氨酶和磷酸肌酸激酶等心肌酶谱含量升高(P<0.05),IL-1和TGF-β相对表达量升高(P<0.05);与单纯照射组比较,蓝莓干预组的苏木精-伊红及Masson染色显示心肌损伤的程度轻,血清谷草转氨酶和磷酸肌酸激酶等含量低,IL-1和TGF-β相对表达量低,相关指标均高于空白对照组(P<0.05)。结论:心肌组织中IL-1和TGF-β表达升高在心肌损伤的发生机制中起到重要作用。在一定剂量范围内,蓝莓花色苷可以降低放射性心肌损伤程度,且随着剂量升高呈现正相关,其机制可能与降低IL-1和TGF-β的表达有关。 Objective:Investigate the changes of IL-1and TGF-βexpression in radiation-induced tissue damage in the heart and the effects of blueberry anthocyanins intervention.Method:40 rats were randomly and equally divided into the control group,irradiation group,20mg/kg blueberries irradiated group and 80mg/kg blueberries irradiated group.They were irradiated by a single dose of whole breast irradiation 15 Gy.Before and after irradiation,blueberry anthocyanins were given to them by qastric lavage everyday,8 days later,detect the myocardial enzymes indicators,myocardial pathological changes by HE and Masson staining observe,analysis the change of the relative expression of IL-1and TGF-βamount by the use of quantitative PCR instrument.Result:Compared with the control group,normal irradiation group showed a certain degree of myocardial damage,myocardial enzymes such as serum aspartate transaminase and creatine kinase elevated(P〈0.05),relative expression levels of IL-1and TGF-βincreased(P〈0.05).Compared with the irradiation group,blueberries intervention groups showed a lesser degree of myocardial injury a decreased level of,Serum aspartate transaminase and creatine kinase(P〈0.05),a relatively low volume of IL-1and TGF-βexpression,higher than the control group(P〈0.05).Conclusion:The elevation of IL-1and TGF-βexpression in myocardial tissue plays an important role in the pathogenesis of myocardial injury,whereas blueberries can reduce the extent of radioactive myocardial injury,which may be associated with the decreased expression of IL-1and TGF-β.
