LPS对人牙周膜细胞Toll样受体4及下游炎症因子表达的影响 被引量:12

摘要 目的:观察革兰氏阴性菌外膜成分脂多糖(LPS)对牙周膜细胞(PDLCs)Toll样受体4(TLR4)及下游炎症因子表达的影响。方法:用浓度为10μg/m L的LPS对牙周膜细胞进行刺激,Western Blot检测牙周膜细胞上TLR4受体表达,同时ELISA检测培养基上清中炎症因子TNF-α、IL-1β的含量。结果:加入LPS后,牙周膜细胞上TLR4蛋白表达增强,炎症因子TNF-α、IL-1β含量增多,且随LPS的刺激时间增加而呈上升趋势。结论:LPS促进PDLCs的TLR4表达,并导致炎症因子分泌增加,TLR4信号通路在牙周炎发病过程中具有重要作用。 Objective:To examine the effect of lipopolysaccharide(LPS) on expression of Toll-like receptor 4(TLR4) and the downstream cytokines in periodontal ligament cells (PDLCs). Method:Expression of TLR4 in PDLCS stimulated by 10μg/mL LPS was detected by Western Blot,and the level of TNF-α,IL-1βin the supernatant were detected by ELISA. Re-sult:In PDLCs stimulated by LPS,the expression of TLR4 increased,and the level of TNF-α,IL-1β increased in a time-dependent manner. Conclusion:Stimulation with LPS could promote the expression of TLR4 and lead to increased secretion of inflammatory cytokines. TLR4 signaling pathway plays an important role in the pathogenesis of periodontitis.
