摘要
目的:探讨8周的有氧跑台运动对D-半乳糖模型阿尔茨海默病(AD)大鼠海马β-淀粉样前体蛋白(APP)和Tau蛋白及其激酶糖原合成酶激酶-3β(GSK-3β)m RNA表达的影响。方法:选用健康雄性8周龄SD大鼠16只,随机分为对照组(C组,n=8),运动组(T组,n=8)。两组大鼠连续8周经腹腔按120mg/kg·d的浓度注射D-半乳糖造模AD大鼠,同时对T组大鼠进行连续8周,每周5次的有氧跑台运动。运用实时荧光定量RT-PCR的方法检测两组大鼠海马APP、Tau蛋白和GSK-3β的m RNA表达水平。结果:8周的有氧跑台运动显著下调了T组大鼠海马APP、Tau蛋白和GSK-3β的m RNA表达水平;其中,APP的下调幅度为42%(P<0.05),Tau蛋白的下调幅度为45%(P<0.01),GSK-3β的下调幅度为38%(P<0.05)。结论:8周的有氧跑台运动有效抑制了APP和Tau蛋白的基因表达水平,其机制可能是与运动抑制了GSK-3β的基因表达水平有关。
Objective: To explore the effects of 8-week aerobic treadmill training on expressions of β-amyloid precursor protein(APP), Tau protein and glycogen synthase kinase- 3β(GSK- 3β) m RNA in hippocampus of D- galactose(D-gal) Alzheimer's disease(AD) rats.Method: Sixteen male Sprague-Dawley(SD) rats were randomly divided into control group(C, n=8), exercise group(T,n=8). All rats of two groups were injected D- gal 120mg/(kg · d) in abdominal for 8 weeks to obtain AD model rats. Group T was given aerobic treadmill, 5 times a week for 8 weeks. The expression levels of APP,Tau and GSK-3β in hippocampus of all rats were inspected by real-time fluorescence quantitative RT-PCR.Result: After 8- week aerobic treadmill exercise made the expressions of APP mRNA(42%, P〈0.05), Tau mRNA(45%, P〈0.01) and GSK- 3β(38%, P〈0.05) were down- regulated significantly in hippocampus of Dgal AD rats.Conclusion: Eight- week aerobic treadmill training could made some inhibition effects on the expressions of APP and Tau m RNA in D-gal AD rats. The mechanisms might be related with the lower expression level of GSK-3β m RNA, which was down-regulated by 8 weeks aerobic treadmill exercise and would made some important regulation effects on the levels of APP and Tau protein expression.
