摘要
试验以苯酚-浓硫酸法测定多糖,微波辅助浸提和乙醇沉淀法对杏鲍菇子实体粗多糖进行提取和分离纯化实验研究。结果表明,杏鲍菇子实体多糖提取的较优工艺条件为提取温度75℃、提取时间12min、料水比1∶35、微波功率400W、提取2次,在此较优条件下杏鲍菇粗多糖的得率达7.348%;纯化工艺条件为用40%氯仿、10%正丁醇、2%甲醇、40%乙醇、0.01%NaCl脱蛋白制精多糖。
The content of polysaccharide in Pleurotus eryngii was determined by phenol- sulfuric acid method. To extract rough polysaccharide from the Pleurotus eryngii's fruit-body, microwave-assisted hot watter extraction and precipitate with alcohol was used. The results show the optimal extraction processing conditions of polysaccharide were extraction temperature of 75 ℃ , microwave time of 12min, material-water ratio of 1:35(g/mL) and microwave power 400W. Under the optimal extraction conditions, the extraction rate of polysaccharide from Pleurotus eryngii's fruit-body was up to7.348%; 40% of chloroform, 10% of 2-butanol, 2% of methanol, 40% of ethanol, 0.01% of NaCl was used to deproteinizating for pure polysaccharide.
