X射线对小鼠胰腺组织结构及血清胰岛素含量和胰淀粉酶活性的影响

Effects of X-ray Irradiation on Pancreas Structure,Serum Insulin and Pancreatic Amylase Activity in Mice

摘要 探讨X射线辐射对小鼠(Mus musculus)胰腺组织的损伤效应,对血清胰岛素含量及胰淀粉酶活性的影响,分析X射线对小鼠胰腺内、外分泌部功能的影响。将成年小鼠随机分为1 Gy、3 Gy、5 Gy实验组和0 Gy对照组(每组20只),各组小鼠给予对应剂量X射线辐射(连续3 d,每天辐射一次,每次3 min)。取胰腺组织制作组织切片,用生物显微技术观察胰腺组织结构的变化,取小鼠血液,制备血清,用双抗体夹心法测定胰岛素含量,用比色法检测胰淀粉酶酶活性的变化。结果显示,X射线辐射引起小鼠胰腺组织肿胀,细胞核变大,细胞变性、坏死;X射线辐射后5~15 d,1 Gy辐射组小鼠胰岛素含量与对照组相比较有不同程度的升高,差异显著或极显著(P〈0.05或P〈0.01),第20天时略有下降,差异不显著。3 Gy辐射组各期小鼠胰岛素含量与对照组相比均下降,差异显著或极显著(P〈0.05或P〈0.01)。5 Gy辐射组各期小鼠胰岛素含量与对照组相比均下降,差异显著或极显著(P〈0.05或P〈0.01),第15天起全部死亡;1 Gy辐射组小鼠胰淀粉酶活性相比对照组变化不显著(P〉0.05),3Gy辐射组小鼠胰淀粉酶活性相比对照组均有不同程度的升高,差异显著或极显著(P〈0.05或P〈0.01);5 Gy辐射组小鼠胰淀粉酶活性较对照组降低,差异极显著(P〈0.01)。结果表明,X射线辐射能引起小鼠胰腺组织结构损伤,影响小鼠血清胰岛素含量和血清胰淀粉酶的活性,引起胰腺功能异常。 To explore the adverse effects of X-ray irradiation on pancreatic tissue structure,serum insulin level and serum pancreatic amylase activity in mice,adult mice were randomly allocated to 0 Gy,1 Gy,3 Gy,and5 Gy groups( 20 mice in each),and each group was exposed to 3 min corresponding dose of X-ray irradiation once a day for 3 days. The changes of pancreas tissue structure were observed by optical microscope,and the changes of serum insulin concentration was determined with double antibody clip art method,and pancreaticamylase enzyme activity in serum was detected with colorimetric method. Results showed that the X-ray irradiation caused pancreatic tissue swelling,with bigger cell nucleus and degenerative necrosis( Fig. 1); The insulin level of 1 Gy irradiation group was higher than that of control group after 5- 10 days( P 〈 0. 05 or P 〈0. 01),but fell slightly by 20 days( Fig. 2). Insulin levels in 3 Gy and 5 Gy irradiation mice decreased compared to control group. 5 Gy mice died by 15 days( Fig. 2). Amylase activity in 1 Gy irradiation group was close to that of control group( P 〈 0. 05)( Fig. 3),while amylase activity in 3 Gy irradiation group was higher than that of control group( P 〈 0. 05 or P 〈 0. 01)( Fig. 3). Amylase activity in 5 Gy irradiation group decreased compared to that of the control group( Fig. 3). These results show that pancreas tissue structure is damaged and serum insulin and pancreatic amylase activity are decreased by X-ray irradiation. X-ray causes abnormal function of the pancreas.
