百合品种‘Siberia’ rDNA ITS序列的克隆及序列比对

摘要 本文对东方百合品种‘Siberia’样品进行ITS的序列测定,将得到的序列与GenBank上百合属42个种的rDNA ITS进行比对,并在此基础上构建了系统发育树。序列比对结果表明东方百合品种‘Siberia’的rDNA ITS与其它种的同源性不高,序列的相似性为70%左右。系统发育树也表明东方百合品种‘Siberia’与同比42个种的同源率较低,证明百合属各种间、种内高度进化,东方百合品种‘Siberia’的形成是经过多世代多次进化而完成。 In order to identify oriental hybrids "Siberia" and its closely related genera, the rDNA ITS sequences of 42 accessions were analyzed,the inter and intraspecific genetic distances were calculated, and the phylogenetic trees of all the accessions tested were constructed based on the obtained distance data. The alignment results showed that the rDNA ITS sequence of oriental hybrids ‘Siberia' was different from all alignment sequences, the similarity of oriental hybrids ‘Siberia' and its related genera was about 70%. The phylogenetic tree also showed that the level of similarity between oriental hybrids "Siberia" rDNA ITS and its related genera was low. The results would provide a reference on identifying the species and genus relationship of Lilium by using ITS sequences.
