摘要
目的 评价中药川芎提取物对高眼压状态下视功能的保护作用.方法 40只家猫随机分为空白组、模型组、川芎嗪组、脑神经生长素对照组,每组10只动物,前房注射20g·L-1甲基纤维素制作家猫急性高眼压模型,前肢静脉穿刺注射给药,于造模后第1天开始,每天1次,连续给药9d,以图形视网膜电图(pattern electroretinogram,PERG)和图形视觉诱发电位(pattern visual evoked potential,PVEP)为评价手段,观察用药前后以上指标的变化.结果 高眼压能够严重损害家猫视神经功能,表现为PERG及PvEP各参数峰潜时明显延长,振幅降低,与术前相比差异具有显著统计学意义(P<0.01).川芎嗪组、脑神经生长素对照组均使高眼压家猫异常的P50、N95波峰潜时缩短,振幅提高,表现为:干预10天后川芎嗪组及阳性药物对照组P50波峰潜时缩短,振幅提高,与模型组比较差异具有统计学意义(P<0.01);P50峰潜时与干预前比较,川芎嗪组及阳性药物对照组P<0.05;P50振幅与干预前比较川芎嗪组及阳性药物对照组均明显提高(P<0.01).干预10天后川芎嗪组N95波峰潜时缩短,振幅提高,与干预前、模型组差异均有统计学意义(P<0.01);川芎嗪组对振幅的改善优于阳性药物对照组(P<0.05);川芎嗪组及阳性药物对照组对高眼压家猫PVEP之P100波变具有改善作用,干预10天后P100波峰潜时明显缩短,2种药物与模型组比较差异具有统计学意义(P<0.01),川芎嗪组作用更明显,与干预前比较差异亦具有统计学意义(P<0.01、P<0.05);干预10天后P100波振幅提高,川芎嗪组及阳性药物对照组较之干预前振幅提高(P<0.01、P<0.05).结论 中药川芎提取物能够使高眼压家猫异常的PERG及PVEP之P50、N95、P100波峰潜时缩短,振幅提高,对高眼压状态下视神经的保护有益.
Objective To evaluate the protective effect of Ligusticum wallichii extraction on retinal ganglion cells under high intraoeular pressure (IOP). Methods Forty cats were randomly divided into normal group,model group,ligustrazine group and cranial nerve growthine group, 10 cats in each group. The high IOP model was established by injecting 20 g ~ L~ cellogran into anterior chamber of cats, the vein in anterior limb was punctured from the first day after modeling,once a day for 9 days. The changes of pattern electroretinogram(PERG) and pattern visual evoked potential (PVEP) were observed before and after puncturation. Results The high IOP damaged the function of optic nerve of cat severely according to the parameter changes of PERG and PVEP, including its peak latency obviously prolonged and amplitude decreased, there was significant difference compared with pre-operative parameters (P ~ 0. 01 ). The ligustrazine group and cranial nerve growthine group both could shorten the peak latency and improve the amplitude of P50 and N95 wave. The peak latency of PS0 wave was shorted and the amplitude was improved in ligustrazine group and cranial nerve growthine group after 10 days, there were significant differences compared with model group(both P〈0.01). Compared with pre-operation, the peak latency of P50 wave in ligustrazine group was significantly decreased (P〈0. 01), the amplitude of P50 wave was improved in ligustrazine group and cranial nerve growthine group(P〈0.01), the protective effects of ligustrazine group were better than those of cranial nerve growthine group(P〈i0.05). The P100 wave of cat with high IOP was improved in ligustrazine group and cranial nerve growthine group, including the peak latency of Pl00 wave was obviously shortened after 10 days,there were significantly differences compared between two drug groups and model group(P^0.01); The amplitude of Pl00 wave was improved after 10 days,especially in ligustrazine group, there was significantly differences compared with pre-operation(P%0.01 ,P^0.05), the improvement in ligustrazine group was better than tb.at in model group and in cranial nerve growthine group (P%0.01,P%0.05). Conclusion The ligustrazine can shorten the peak latency and improve the amplitude of P50, N95 and Pl00 wave of abnormal PERG and PVEG in cat with high IOP, which may be benefit to the protection of optic nerve under high IOP.
