炙甘草汤减缓兔右心房颤所致心肌纤维化作用

Study the effection of Zhigancao Decoction on reducing myocardial fibrosis in right atrial fibrillation New Zealand rabbits

目的研究炙甘草汤逆转快速起搏兔右心房(RAP)诱导心房颤动(AF)致右心耳心肌心肌纤维化的作用。方法新西兰大白兔动物模型随机分为假手术组(A组):植入电极不行快速起搏;起搏组(B组):行短期快速起搏兔右心房12h致AF模型;炙甘草汤水煎液阴性方组(C组):给予炙甘草汤阴性方水煎液灌胃2次/d,连续30d后行短期快速起搏兔右心房12h致AF模型;炙甘草汤组(D组):给予炙甘草汤水煎液灌胃2次/d,连续30d后行短期快速起搏兔右心房12h致AF模型;每组实验动物各8只。建立房颤兔模型,行兔右心耳Masson染色和免疫组化方法检测基质金属蛋白酶-9(MMP-9)蛋白表达。检测右心耳组织场电位时程(fAPD),并观察炙甘草汤对右心耳fAPD的作用。结果成功建立快速起搏右心房诱导房颤兔模型,Masson染色显示B组和C组心肌纤维化明显增加,D组心肌纤维化减弱。MMP-9结果显示,B组和C组MMP-9呈强阳性,D组呈弱阳性。B组和C组fAPD明显缩短,差异有统计学意义(P<0.05)。给予炙甘草汤水煎液之后,可作用于房颤兔右心耳组织fAPD导致fAPD延长(P>0.05)。结论炙甘草汤可逆转房颤兔心肌纤维化,同时缩短fAPD的时程。

Objective To study the Zhigancao Decoction reducing right atrial myocardium of myocardial fibrosis based on rapid atrial pacing（RAP）in rabbits.Methods New Zealand white rabbits were randomly divided into sham operation group（A group）with implanted electrodes,no rapid pacing;pacing group（B group）with AF model induced by short-termrapid pacing rabbits right atrial 12h;negative prescription of Zhigancao Decoction group（C group）with negative prescription of Zhigancao Decoction,Decoction orally 2times/d for 30 d,continuously then short-term rapid pacing of right atrial 12 hinduced AF model;Zhigancao Decoction group（D group）with Zhigancao Decoction orally 2times/d for 30 d,continuously then short-term rapid pacing of right atrial 12 hinduced AF model.experimental animal of each group with 8rats.Atrial fibrillation model established in rabbits.atrial myocardial fibrosis was tested by Masson staining and protein expression of matrix metalloproteinase-9（MMP-9）was detected with pathologic immunohistochemistry.right atrial appendage tissue field action potential duration（fAPD）was measured by microelectrode arrays（MEA）.Results the rapid atrial pacing successfully inducted atrial fibrillation model in rabbit.Masson staining showed myocardial fibrosis significantly increased in B and C groups,myocardial fibrosis reduced in D group.protein expression of MMP-9was strongly positive in B and C group,protein expression of MMP-9was weakly positive in D group.fAPD significantly decreased in B group and C group,the difference was statistically significant（P〈0.05）.After administration of Zhigancao Decoction,fAPD prolonged（P〉0.05）.Conclusion Zhigancao Decoction can reduce myocardial fibrosis of atrial fibrillation in rabbits,and short the duration of fAPD.
