摘要
以聚合物纳米微球为层析介质,采用HPLC检测方法,开发高纯度棘白霉素B母核的分离纯化工艺,并对层析条件进行优化。结果表明,当选择Uni PS 30型聚合物纳米微球为层析介质、上样量为1.2g·(100g介质)-1、以体积分数为3%的乙醇进行洗脱、流速为12mL·min-1时,得到的棘白霉素B母核纯度大于98%。建立了高纯度棘白霉素B母核的分离纯化工艺,流程简单、可行,为该产品产业化开发提供了一定的理论基础。
Using polymer nanoparticles as the chromatography medium,a method for separation and purification of high-purity echinocandin B nucleus was developed.The purity was detected by HPLC,the chromatographic conditions were optimized.Results showed that,when selecting polymer nanoparticles Uni PS 30 as the chromatography medium,loading capacity was 1.2g·(100g medium)^-1,volume fraction of ethanol was 3%,and the flow rate was 12mL·min^-1,echinocandin B nucleus with the purity of 98% were obtained.The method for separation and purification of echinocandin B nucleus was established.It was simple and feasible,which provided a theoretical basis for the industrialization of echinocandin B nucleus.
