摘要
目的研究益气化痰通络法对博来霉素诱导的大鼠肺纤维化的干预作用,以及对血清中转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)表达的影响。方法雄性Wistar大鼠90只,随机分成对照组15只,模型组75只,模型组经博来霉素造模后,随机分成中药组、激素组、乙酰半胱氨酸组、罗红霉素组和生理盐水组(模型对照组)各15只,正常鼠粮喂养并每日给药,分别于第15日、30日和45日后处死大鼠,对大鼠肺组织做病理切片HE染色,并用ELISA法检测血清中TGF-β1、TNF-α的表达。结果 HE染色示中药组的肺泡炎症、肺纤维化程度较模型对照组显著减轻,中药组大鼠血清中TGF-β1、TNF-α的表达显著低于模型对照组,差异具有统计学意义(均P<0.05或P<0.01)。结论益气化痰通络法对博来霉素诱导的大鼠肺纤维化具有干预作用,其机制可能为减少血清中TGF-β1、TNF-α的表达。
Objective:To study the intervention effect of Yiqihuatantongluo Decoction on rats with pulmonary fibrosis induced by bleomycin and impact of it on serum TGF-β1 and TNF-αlevel.Methods:90 male Wistar rats were randomly divided into control group(15 cases) and model group(75 cases) induced by bleomycin.Then,the model group were again randomly divided into traditional Chinese medicine group,hormone group,acetylcysteine group,roxithromycin group and normal saline group (model control group),with 15 cases in each group.Normal food and corresponding drugs were given daily.The rats were partially killed after 15th d and 30th d,and all killed after 45 d.Lung tissue was made pathological section and HE staining.ELISA method was used to detect the expressions of TGF-β1 and TNF-α in serum.Results:HE staining showed that alveolar inflammation and the degree of pulmonary fibrosis of rats in the traditional Chinese medicine group reduced significantly,and the expressions of TGF-β1,TNF-αin the serum of Chinese traditional medicine group are significantly lower than those of model group.Conclusion:Yiqihuatantongluo Decoction has intervention effect on rats with pulmonary fibrosis induced by bleomycin,and its mechanism may be related to reducing the expressions of TGF-β1 and TNF-αin serum.
