补阳还五汤联合脂肪来源干细胞移植对脑缺血大鼠神经功能的影响

Effect of Buyanghuanwu Decoction Combined with Adipose-derived Stem Cells Transplantation on Nerve Function in Experimental Rats with Cerebral Ischemia

摘要 目的探讨补阳还五汤联合脂肪来源干细胞(ADSCs)移植对脑缺血损伤大鼠的神经保护作用。方法采用线栓法制备大鼠脑缺血模型,大鼠随机分为假手术组、模型组、ADSCs移植组(移植组)、补阳还五汤联合ADSCs移植组(联合组)。各组又分7、14、21 d 3个时间点的亚组,观察各组大鼠不同时间点的神经功能评分、脑梗死面积及脑组织病理形态学变化。结果与模型组比较,移植组、联合组大鼠造模后14、21 d神经功能评分均显著降低(P<0.05或P<0.01),且联合组大鼠21 d神经功能评分明显优于移植组(P<0.05);各时间点移植组与联合组大鼠脑梗死面积明显小于同时相的模型组(P<0.01),且14、21 d联合组优于移植组(P<0.05);造模后HE染色显示模型组大鼠脑组织缺血周边神经元数量减少,细胞明显水肿,治疗后脑组织损伤明显减轻,联合组的病理损伤最轻。结论补阳还五汤联合ADSCs移植能减轻脑缺血后神经功能损伤,具有脑保护作用。 Objective: To explore the protective effects of Buyanghuanwu Decoction combined with adipose-derived stem cells (ADSCs) transplantation on cerebral ischemia in rat. Methods: Rats were randomly divided into the sham operation group,the model group,the ADSC transplantation group,the Buyanghuanwu Decoction combined with ADSCs group. Cerebral ischemia animal models were established with nylon thread. Each group was divided into 3 sub-groups according to 3 time points including the 7 d,the 14 d and the 21 d. Some indicators were observed in each group at different time points such as neurological function,cerebral infarction size and brain issue morphological changes. Results: Compared with model group, the neurological score were significantly decrease in transplantation group and combination group at 14 d and 21 d after MCAO(P〈 0.05 or P〈 0.01 ). This index in the combination group was better than that in the transplantation group at 21 d (P〈 0.05). The infarction area in transplantation group and combination group were significantly decreased compared with model group at the same time-phase. The size in the combination group was better than that in the transplantation group at 14 d and 21 d(P〈 0.05). After MCAO, HE staining showed injuried never cells with twisted axonal processes and cells loss were evident in the ischemia penumbra , and those changes were very few in each treatment group especially the combination group. Conclusion: Buyanghuanwu Decoction combined with ADSCs transplantation can improve cerebral ischemic injury neurological recovery. It has cerebral protective effect.
