摘要
目的 观察附子及附子、干姜配伍对H2O2诱导的乳鼠心肌细胞抗氧化作用.方法 取1~3 d乳鼠的心肌细胞,常规培养,观察心肌细胞形态学,以免疫组化进行心肌细胞纯度鉴定.实验分为对照组、模型组、附子水煎液组20 mg/mL,附子干姜水煎液组20 mg/mL,预处理2h后,用1 mmol/LH2O2氧化损伤2h,检测培养液中心肌细胞跳动频率,丙二醛(MDA)及超氧化物歧化酶(SOD)活性.结果 与模型组相比,用附子水煎液和附子干姜水煎液预处理后的心肌细胞跳动频率增加(P<0.01),附子干姜水煎液组SOD活性明显提高(P<0.01).结论 附子干姜水煎液对H2O2诱导的心肌细胞损伤具有一定的保护作用.
Objective:To investigate the antioxidant effect of aconite and compatibility of aconite dried ginger pretreatment on neonatal rat myocardial cells induced by H2O2.Methods:Myocardial cells were clipped from 1~3 days of neonatal rat.Cell morphologic change was observed under an inverted microscope and purity identification of myocardial cells were studied by immunohistochemistry.Different groups were set after conventional cultur,such as normal groups,the model groups,20 mg/kg aconite decoction groups,20 mg/kg aconite and dried ginger decoction.After pretreatment 2 hours and the 1 mmol/L H2O2 oxidative injury 2 hours.The different groups were observed about cell pulsating.The activity of MDA and SOD was detected.Results:Compared with model groups,in the aconite and dried ginger decoction pretreatment group,the beating rate of the myocardial cell was increased (P 〈 0.01).The SOD activity were also improved (P 〈 0.01).Conclusion:It has the protective effect of the aconite and dried ginger decoction on myocardial injury induced by the H2O2.
