摘要
目的探讨辣椒素及二氢辣椒素对大鼠神经胶质瘤C6细胞、骨肉瘤ROS 17/2.8细胞的抑制作用,C6细胞凋亡率及细胞内氧自由基(ROS)的影响。方法 CCK-8法检测辣椒素及二氢辣椒素(50、100、150、200μmol/L)对大鼠神经胶质瘤C6细胞的抑制作用;Annexin V-FITC、PI双染检测辣椒素及二氢辣椒素对C6细胞凋亡率的影响;DCFH-DA检测C6细胞内ROS水平。结果辣椒素及二氢辣椒素显著抑制C6细胞、ROS 17/2.8细胞的增殖,且与时间和药物浓度呈正相关。辣椒素处理C6细胞12 h后,早期凋亡率明显增加,ROS水平显著提高。结论辣椒素及二氢辣椒素对大鼠神经胶质瘤C6细胞、骨肉瘤ROS 17/2.8细胞具有显著抑制作用,其抑制C6细胞增殖机制与促进肿瘤细胞凋亡,增加ROS的产生密切相关,辣椒素及二氢辣椒素有望成为新型抗肿瘤药物。
Objective To study the inhibition rate of capsaicin (Cap) and dihydrocapsaicin(DHC) on C6 glioma cells and ROS 17/2.8 osteosarcoma cells, and to observe the apoptosis rate and ROS of Cap/DHC on C6 glioma cells. Methods CCK-8 assay was used to detect the inhibition rate of Cap/DHC(50, 100, 150 and 200 μmol/L) on C6 glioma cells. The effect of capsaicin and dihydrocapsaicin on the apoptosis rate of C6 glioma cells were tested by Annexin V-FITC and double staining methods, while ROS expression of C6 cells was by DCFH-DA. Results Cap and DHC presented significant inhibitory effects on proliferation of C6 cells and ROS 17/2.8 cells and were positively correlated with time and drug concentration. After treated with 200 μmol/L Cap for 12 h, the early apoptosis rate of C6 cells increased obviously. The level of ROS improved significantly. Conclusion Cap and DHC showed obvious inhibition against C6 glioma cells and ROS 17/2.8 osteosarcoma cells. The inhibition mechanism of C6 cells was related with promoting tumor ceils apoptosis and increasing ROS formation. Cap and DHC may become new potent anti-cancer drugs.
